XYZ belongs to Lindera genus in the family Lauraceae which has the functions of warming channels,regulating vital energy and treating stomach pain and gastric ulcer.It is mainly distributed in Hunan,Hubei,Yungui,Guangxi and Sichuan in China.WY belongs to Lindera genus in the family Lauraceae which is traditionally used to warm stomach,regulate vital energy and relieve pain.In additional,it has extensive pharmacological activities in digestive system,cardiovascular system and central nervous system.In this paper,the potential antidepressant activities and mechanisms of ethanol extracts of XYZ and WY were preliminarily evaluated.The chemical constituents of L.fragrans leaves were isolated,purified and identified.In order to explore the effects of different doses of XYZ and WY crude extracts on the depressive behavior by CUMS,we observed the changes of weight,sucrose preference and tail suspension immobility time.Then the changes of serum corticosterone,5-hydroxytryptamine and norepinephrine in hippocampus of mice were detected to explore their mechanisms.The results showed that the extracts of XYZ and WY can significantly increase the sucrose preference and reduce the immobility time in tail suspension test in depressive mice.The antidepressant effect of XYZ and WY may be related to its regulation of 5-hydroxytryptamine,norepinephrine in hippocampus and corticosterone in serum.The air-dried leaves of L.fragrans(2.9 kg),after comminution,extract with 95% ethanol 3 times.Three parts were obtained though extracted by petroleum ether,ethyl acetate and n-butanol,respectively.The petroleum ether and ethyl acetate soluble part were separated by silica gel column chromatography,MCI,Sephadex LH-20 and HPLC and twenty compounds were obtained.Finally,fifteen compounds were identified as: Nordicentrine(1),Phanostenine(2),Cryptodorine(3),Neolitsine(4),Dehydroneolitsine(5),Anolobin(6),Kaempferol-3-O-?-L-rhamnoside(7),Kaempferol 3,7-O-?-L-dirhamnoside(8),Kaempferol 3-O-?-D-xylopyranosyl-(1?2)-?-L-rhamnopyranoside(9),p-Methoxyphenylacetic acid(10),4-Methoxycinnamic acid(11),Demethylmacrosporine I(12),Daucosterol(13),Stigmasterol(14),?-Sitosterol(15).
