Aim: Clinical investigation showed that diabetes was associated with stroke and worsened brain damage and prognosis after stroke.Diabetes have effect on the occurrence and prognosis of stroke through destroying the cerebral vessels.The clinical effect of fasudil mainly act on vascular disease,we want to study the effects of fasudil on the prognosis of diabetic stroke by chronic pretreatment.Methods: 250-300 g Sprague-Dawley rats were randomly divided into nondiabetic stroke group,diabetic stroke group and fasudil pretreatment group.The diabetes group rats treated with intraperitoneal injection of streptozotocin(60 mg/kg),same dose of citrate buffer inject into control group rats.Fasudil group rats receives daily fasudil intraperitoneal injection at 10 mg/kg for three consecutive weeks.4 weeks later,all the experimental groups rats treated middle cerebral artery occlusion for 90 minutes.After sacrifice,the brain sample was collected for following experiments,infarct volume,edema volume,blood brain barrier were detected by immunohistochemistry.Inflammatory factors were examined by RT-PCR using tissue RNA.Results: The concentration of blood glucose at 15 mmol/L or above proved that the diabetes model was successful.Fasudil pretreatment decrease stroke mortality of diabetes from 43.75% to 31.25%,the infarction decrease volume from 52.95% + 12.7% to 45.97% + 6.7%.Gap formation of tight junction and IgG leakage were reduced(p<0.05).The inflammatory factors expression decrease(p<0.05,p<0.01)in Fasudil pretreatment after diabetic stroke.Conclusion: Diabetes aggravate the mortality of cerebral ischemia rats.The long-term pretreatment of Fasudil can reduce mortality of diabetic stroke,decrease cerebral infarction volume and inflammatory factors expression,and protect the blood-brain barrier.
