Objectives: To investigate the molecular mechanism of CC motif chemokine ligand 19(CCL19)in suppressing colorectal carcinoma cell proliferation.Methods: Lentiviral vector was used to build stable transfected colorectal cancer cell line SW1116-CCL19 and SW480-CCL19.CCK-8 cell proliferation assay was used to determine the relative cell growth.Agar clone formation assay was used to confirm the suppress effect of CCL19.Cell cycleand apoptosis were analyzed by flow cytometry..To confirm the impact of CCL19 on colorectal carcinoma in vivo,we generated xenograft models by implanting SW1116-CCL19(stably expressing CCL19)or the control SW1116 vector cells into nude mice.Western blot was used to detect the relationship between CCL19 and ?-catenin expression levels in SW1116 and SW480.Results: Compared with negative control group and blank control group,overexpressing CCL19 suppressed proliferation by arresting SW1116 and SW480 in G1-phase.Western blot indicated that ?-catenin and cyclinD1 expression were inhibited in overexpressing group.In vivo,overexpressing CCL19 inhibited tumor growth compared with negative control.In CRC tissues,Expression of ?-catenin is negatively related to CCL19.Conclusion: The results suggests that CCL19 inhibited cell proliferation by inhibiting expression of Wnt/?-catenin pathway.
