Objective:Through the intervention of Mongolian medical osteopathy and matrix metalloproteinase inhibitors in lumbar degenerative rats,Explain the mechanism of degeneration of intervertebral disc tissue by two methods from the perspective of morphology,and from the genetic level to confirm the efficacy of traditional Mongolian traditional bone manipulation on intervertebral disc degeneration.Methods:40 SPF(Specefic pathogen Free,SPF)8-week-old SD(Sprague-Dawley)rats were randomly divided into four groups:manual group,tetracycline group,model control group,and blank group(n=10).The manual group,the tetracycline group,and the model group were used to construct a lumbar degenerative animal model using acupuncture annulus.After successful modeling,the opponent group was treated with Mongolian medicineTraditional massage technique treatment for 2 weeks,the tetracycline group was injected with subcutaneous four-ring injection for 2 weeks,and the model control group was injected with normal saline for 2 weeks.Blank group is not treated.After modeling for 6 weeks,the X-ray examination,pathological examination and the expression of COL-2a,COL-la and Aggrecan genes in the lumbar spine were detected in each group of experimental animals.Results:The lumbar spine X-ray and pathological examination in the blank group showed no significant change before modeling.In the model control group,the anterior border of the vertebral body was hyperplastic and calcified.The biopsy showed that the local annulus fibrosus structure disrupted the annulus fibrosus gap widening and the rupture was further aggravated.The tetracycline group compared with pre-treatment,Imaging examination showed no significant change in intervertebral space stenosis,slice examination showed nucleus pulposus nodular hyperplasia with angiogenesis.Manipulative group scoliosis improvement,Intervertebral space stenosis is significantly improved compared with before treatment,slice examination showed nucleus pulposus cell proliferation with angiogenesis.Real-time PCR for intervertebral disc tissue of all experimental animals.Expression levels of COL-2a;COL-la and Aggrecan were detected.The control group and the blank group were statistically significant,(P<0.05).The expression levels of the three genes in the manipulation group and the tetracycline group were statistically significant compared with themodel control group,(P<0.05).The manual group compared with the tetracycline group,the expression of COL-la gene was not statistically significant,(P>0.05).The manual group compared with the tetracycline group,the expression of COL-2a gene was not statistically significant,(P>0.05).In the Aggrecan gene expression assay,the comparison between the manipulation group and the tetracycline group was statistically significant,(P<0.05).Conclusion:Both treatments have the same effect in morphology and pathology,Among the three groups of genes that produce changes in lumbar degeneration,Mongolian medical bonesetting has an effect on the expression of COL-2a,COL-la and Aggrecan in lumbar disc tissue,and the expression of Aggrecan is most significant,it may be one of the mechanisms of this method for the treatment of lumbar degenerative changes.
