Objective To determine the levels of brain-derived neurotrophic factor(BDNF)in hippocampus of rats with depression and serum tumor necrosis factor(TNF-?),interleukin-6(IL-6),and interleukin-1(IL-1).To study the effect of Baihe Dihuang Decoction and Suanzaoren Decoction on hippocampus and inflammatory factors in rats with depression and its antidepressant mechanism.Methods Sixty male Wister rats were selected as experimental subjects.The rats were used to make a rat model of depression using chronic stress combined with isolation.Sixty male rats were randomly divided into blank group,model group,fluoxetine hydrochloride group,Baihe Dihuang decoction and Suanzaoren decoction high,medium and low dose groups,a total of 6 groups.The experiment used a 21-day model of simultaneous modeling and administration.The rats in the other groups except the blank group and the model group were treated with intragastric drugs,the blank group was given distilled water,and the model group was administered with normal saline.The general condition and body weight of the rats were observed during the experiment,and the behavioral tests were performed on the rats using the "open box experiment".On the 22 nd day of the experiment,all the rats were sacrificed,the carotid artery was taken for blood,the plasma was separated,and the contents of TNF-?,IL-6 and IL-1 in the serum of the rats were determined by elisa method;and the hippocampus of the rat brain was quickly taken out.Part of it is used for HE,after fixation,the histopathological changes of hippocampus were observed by light microscopy.In another part,WB(protein immunoblotting)was used to measure the content of BDNF in hippocampus.Results 1.Compared with the blank group,the weight growth rate of the depression model group was significantly slowed down and the number of horizontal movement grids and standing times in the open box test was significantly reduced(P<0.01).Compared with the model group,the weight of the rats in the high dose group of Baihe Dihuang decoction and Suanzaoren decoction and the fluoxetine hydrochloride group increased significantly and the number of horizontal movement grids and standing times in the open box test increased significantly(P<0.01).Compared with the fluoxetine hydrochloride group,the weight gain was significantly increased in the high-dose group of Baihe Dihuang Decoction and Suanzaoren Decoction and the number of moving lattices and the number of standings in the open box test increased significantly(P<0.05).2.Compared with the blank group,Significant pathological changes occurred in the hippocampal tissue structure of the model group,and the nerve cells were deformed or lost,the levels of TNF-?,IL-6 and IL-1 in the serum of the depression model group were significantly increased and the content of BDNF in the hippocampus was significantly decreased(P<0.01).Compared with the model group,Baihe Dihuang decoction and Suanzaoren decoction's high,middle dose group and fluoxetine hydrochloride group can protect nerve damage to varying degrees,the secretion of TNF-?,IL-6 and IL-1 in the serum of Baihe Dihuang decoction and Suanzaoren decoction's high,middle dose group and fluoxetine hydrochloride group was significantly decreased and the content of BDNF in hippocampus was significantly increased(P<0.01).Compared with the fluoxetine hydrochloride group,the secretion of TNF-? in the high dose group of Baihe Dihuang Decoction and Suanzaoren Decoction decreased significantly(P<0.05),the secretion of IL-6 and IL-1 decreased significantly(P<0.01),and the content of BDNF in the hippocampus increased significantly.(P<0.05).Conclusion Chronic unpredictable mild stress(CUMS)combined with isolation orphans can cause weight loss and behavioral activity in rats.Chinese medicine Baihe Dihuang decoction and Suanzaoren decoction have the functions of nourishing yin and clearing heat,nourishing liver and kidney,nourishing the heart and calming the nerves.By adjusting the excessive secretion of TNF-?,IL-6 and IL-1 in serum and the content of BDNF in the hippocampus,Chronic stress combined with the behavioral behavior of isolated orphaned rats,which may be one of the antidepressant mechanisms of Baihe Dihuang decoction and Suanzaoren decoction.
