Objective To investigate the therapeutic effect of human umbilical cord mesenchymal stem cells on early lung and spleen injury in rats with severe burn,and to provide a new method for the treatment of organ injury after severe burn.Methods 72 male Wistar rats were randomly divided into sham injury group,burn group and burn stem cell treatment group.(1)Severe burn model was established:1%pentobarbital sodium 40mg/kg abdominal anesthesia,depilation for skin,94?,After 12 s,30%of the total surface area of the back was burned by(TBSA)?degree scalding in rats.(2)Treatment scheme:the stem cell group was injected with 2×10~6/1mL hUCMSCs,burn through tail vein and the 1mL PBS.(3)Index was detected by pseudo-injury injection:the lung and spleen tissues were collected 24 and 48 hours after injury in each group.Immunohistochemical staining was used to detect the infiltration of neutrophils and macrophages in the lung and spleen.HE staining was used to observe the pathological changes of the two tissues.Tunel staining was used to detect the apoptosis of lung and spleen cells.Elisa was used to detect the lung.And spleen proinflammatory factor TNF-?,Expression of IL-1?and IL-6.The data were tested by variance analysis and SNK-q test.All data were expressed by(x?s).P<0.05was statistically significant..Results The human umbilical cord mesenchymal stem cells can significantly decreased the infiltration of inflammatory cells and the levels of pro-inflammatory factors TNF-?,IL-1?and IL-6 in lung and spleen tissues of rats with severe burn injury.The pathological changes of lung and spleen and the number of apoptosis were significantly decreased in severe burn rats(P<0.05).Conclusion The human umbilical cord mesenchymal stem cells can effectively reduce the infiltration of inflammatory cells in lung and spleen,reduce the inflammatory reaction and pathological changes of injured tissues,reduce the number of apoptosis,and effectively reduce the levels of pro-inflammatory factors TNF-?,IL-1?,IL-6.And then play an effective role in the treatment of early lung and spleen injury in rats with severe burn.
