Objective To detect the demethylation of MAGE-A1,A2 and A3 in oral mucosal hyperplasia,and to analyze the relationship between demethylation status and clinico-pathological features of oral mucosal hyperplasia,,and further analyze the three The correlation between methylation status and pathological degree provides new evidence for the selection of MAGE-A1~A3 as a marker of potential malignant transformation of oral mucosa,and also provides new ideas and new methods for the accurate treatment of oral mucosal hyperplasia.Methods The pathological tissues of the Department of Oral Mucosa,Oral and Maxillofacial Surgery,and Oral and Maxillofacial Head and Neck Surgery of Inner Mongolia Medical University were collected from 2016 to 2018.The pathological diagnosis was clear.Methyl-specific PCR was used to detect the demethylation status of MAGE-A1,A2 and A3 in different pathological stages of oral mucosal abnormal proliferation.Statistical analysis was performed using SPSS 19.0 statistical software.Results 1.Analysis showed that the degree of MAGE-A1 demethylation was aggravated with the severity of pathological grade(p<0.05),and there was a positive correlation.The difference was statistically significant,with gender,clinical manifestations,size and diameter,location of disease,length of disease.There was no significant correlation between smoking and drinking,and there was a positive correlation with age(p<0.05);2.The analysis showed that the degree of demethylation of MAGE-A2 was aggravated with the aggravation of pathological grade(p<0.05),and there was a positive correlation.The difference was statistically significant,with the patient's gender,age,clinical manifestations,size and diameter,and location of the disease.There was no significant correlation between the length of the disease,whether smoking,and whether or not drinking;3.The analysis showed that the degree of demethylation of MAGE-A3 wasaggravated with the aggravation of pathological grade(p<0.05),and there was positive correlation.The difference was statistically significant,and the gender,age,clinical manifestation,size and diameter of the patient,and the location of the disease.There was no significant correlation between the length of the disease,whether smoking,and whether or not drinking.Conclusion 1.MAGE-A1 demethylation modification is involved in the development and progression of oral mucosal dysplasia.The positive rate of demethylation is positively correlated with pathological grade,and the gender,clinical manifesta-tions,size and diameter of the patient,disease course,whether smoking,whether drinking or not,is positively related to age;2.MAGE-A2 and A3 demethylation modification is involved in the occurrence and development of abnormal hyperplasia of oral mucosa.The demethylation positive rate is positively correlated with pathological grade,and has nothing to do with the gender,age,clinical manifestation,size and diameter,site of onset,course of disease,smoking or drinking.3.The combined test MAGE-A1-A3 has more clinical significance than the single test.
