Objective:To investigate the effect of Yiqi Huoxue Jiedu decoction on autophagy in obstructive nephropathy,we observed the expression of autophagy associated gene 5?Atg5?,Beclin-1 and microtubular-associated protein 1 light chain 3?LC3?,and detecting the related single channels of serum and glucocorticoid-induced protein kinase 1?SGK-1?,phosphorylated extracellular signal-regulated protein kinase?P-ERK1/2?and phosphorylated mammal target of rapamycin?P-mTOR?which were regulated.Methods:Totally 48 healthy clean male Wistar rats were randomly divided into sham-operation group?Sham group?,unilateral ureteral obstruction group?UUO group?,valsartan group?Val group?and traditional Chinese medicine group?TCM group?.The animal models were established with unilarteral urteral obstruction?UUO?except for the Sham group.The rats in the Val group and TCM group were treated with valsartan(10mg·kg-1·d-1)and Yiqi Huoxue Jiedu decoction(14g·kg-1·d-1)respectively.The obstructed kidneys were collected at the 10th after UUO.The expression of Atg5,Beclin-1,LC3,SGK-1,P-ERK1/2,P-mTOR were detected by Western blot and immunohistochemistry.Result:1 The expression of Atg5,Beclin-1,LC3,P-mTOR in rats by immunohistochenical methodBoth Atg5 and Beclin-1 were expressed weakly in the Sham group,which were observed in the cytoplasm of renal tubular epithelial cells.In the UUO group,the expressions were increased obviously.In the Val and TCM groups,the expressions were both decreased obviously.LC3 was expressed weakly in the Sham group,mainly in the cytoplasm and cell membrane of renal tubular epithelial cells.In the UUO group,the expressions were increased obviously,mainly in the cell membrane of renal tubular epithelial cells.In the Val and TCM groups,the expressions were both decreased obviously.The expression of P-mTOR in the Sham group was increased obviously,which were observed in the cell nuclear of renal tubular epithelial cells.In the UUO group,the expressions were decreased obviously.In the Val and TCM groups,the expressions were both increased obviously but less than that in the Sham group.2 The expressions of Atg5,Beclin-1,LC3,SGK-1,P-ERK1/2,P-mTOR in rats by Western blotCompared with the Sham group,the expressions of Atg5 and P-ERK1/2in the UUO group were up-regulated obviously?P<0.01?,but in both Val and TCM groups were down-regulated?P<0.01?.Compared with the Sham group,the expressions of Beclin-1,LC3,SGK-1 in the UUO group were up-regulated obviously?P<0.01?,but in both Val and TCM groups were down-regulated?P<0.05?.The expression of P-mTOR was expressed weakly in the UUO group,it was up-regulated obviously in the Sham group?P<0.05?,compared with the UUO group,both Val and TCM groups were increased obviously?P<0.01?,but less than that in the Sham group.Conclusion:1 Autophagy can be activated in the obstructive nephropathy that made by UUO,and the progressive of autophagy was consistented with the activation trend of Mineralocorticoid receptor?MR?.2 Yiqi Huoxue Jiedu decoction can inhibit the activation of MR to correct the abnormality of autophagy in obstructive nephropathy by regulating SGK-1,P-ERK1/2 and P-mTOR pathways.
