Objective To study the effect of cofilin expression on the phenotype and skeleton of rabbit chondrocytes.Methods The knockdown models and over-expression models of cofilin were constructed by siRNA and plasmid transfection respectively.The cytoskeletal and cytoskeleton-related proteins in chondrocytes were detected to study the effect of cofilin on the phenotype and skeleton of chondrocytes.Results The cytoskeleton of chondrocytes became more compact and the actin density in the cell extension direction was denser than that of the control group after cofilin knockdown.The expression of COL?was slightly increased(137.42% ? 25.14%),the results were not statistically significant(P>0.05),but COL?expression was significantly decreased(19.96% ? 14.59%),it was statistically significant(P<0.05).In contrast,the immunofluorescence intensity of the skeleton in cofilin over-expressed chondrocytes was slightly darker and the distribution was slightly wider than that of the control group,the expression of COL?was decreased(44.47%,? 21.67%),the results were statistically significant(P<0.05),while the expression of COL ? was slightly increased(116.16%? 13.18%),it was not statistically significant(P>0.05).Conclusion The cofilin plays an important role in the chondrocytecytoskeleton;The decrease of cofilin expression could lead to a decrease expression of COLII(a characteristic protein of chondrocytes)and the dedifferentiation tendency of chondrocytes;The increase of cofilin expression could reduce the expression of COLI and prevent chondrocytes from differentiation;The expression of cofilin plays an important role in chondrocytes on maintaining chondrocytes normal phenotype and properties.
