OBJECTIVE: Long non-coding RNA(lncRNA)is dysregulated in various tumors and plays a regulatory role in epigenetics,transcription,post-transcription,and translation.This experiment detected the expression of lncRNA MEG3,XIST and ZFAS1 in human pituitary adenoma(PA),and analyzed its possible association with clinical indicators,and further explored its relationship with the expression of P53 and Bcl-2 in pituitary adenomas.It is speculated that its possible mechanism provides theoretical support for the early diagnosis and targeted treatment of Invasive pituitary adenoma.METHODS: From November 2017 to June 2018,43 patients with PA from the Department of Neurosurgery,Qingdao University Hospital were included in our study,divided into invasive pituitary adenomas group and non-invasive pituitary adenomas group.The expression levels of lncRNA MEG3,XIST and ZFAS1 were detected by RT-PCR,The expression levels of protein P53 and Bcl-2 were detected by western blot.Then analyzed the correlations between the expression level of lncRNA and protein expression levels,and clinical data.Results: 1.The expression level of XIST in IPA tissues was significantly higher than that of non-invasive PA(P<0.05),and its expressions were also related to gender.In functional PA,the expression level of MEG3 in non-invasive PA was significantly higher than that in invasive one(P<0.05).The expression level of ZFAS1 in IPA tissues had no significant difference with that of non-invasive PA(P>0.05).2.The expression of P53 was significantly lower in IPA(0.27±0.13)than in non-invasive PA(0.45±0.15)(P<0.05),and the expression of Bcl-2 was significantly higher in IPA(0.51±0.29)than in non-invasive PA(0.19±0.09)(P<0.05).3.In PA tissues,the expression level of XIST correlated negatively with P53 expression(R=-0.476,P<0.05),and correlated positively with Bcl-2 expression(R=0.554,P< 0.05);The expression of MEG3 correlated positively with P53 expression in functional PA tissues(R=0.718,P<0.05),and had no significant correlation with Bcl-2 expression(R=0.175,P>0.05).Conclusion: 1.XIST may play a carcinogenic role in the occurrence and development of pituitary adenomas by regulating the expressio n of P53 and Bcl-2.2.MEG3 may play the role of tumor suppressor through P53 pathway in functional pituitary tumors.3.The expression of XIST and MEG3 may be of great value in judging the invasion of pituitary adenomas,and have important prospects in the research of targeted therapy.
