| Objectives Toll-like receptor 4(TLR4),a key regulator of innate and acquired immunity,is associated with the development of various cancers.However,the relationship between TLR4 gene polymorphism and susceptibility to esophageal cancer has rarely been reported.The purpose of this study was to investigate the relationship between genetic variants of TLR4 gene and the risk of esophageal cancer in Chinese population,and to screen potential susceptibility biomarkers for esophageal cancer.MethodsIn this study,a total of 480 cases of esophageal cancer were collected as the case group and 480 healthy subjects as the controls.Three SNPs of TLR4(rs1927914,rs11536891,rs7873784)were selected as research targets after analyzing the database.Using polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP)and Taqman SNP Genotyping Assay respectively.χ~2 test was used to compare the distribution differences of relevant factors such as gender,age and smoking status.The odds ratio(OR)and 95%confidence interval(95%CI)were calculated by unconditioned Logistic regression model to evaluate the relationship between the mutation of three SNPs genes and the risk of esophageal cancer.ResultsCompared with TLR4 rs1927914 AA genotype carriers,the risk of esophageal cancer was significantly reduced in rs1927914 GG genotype carriers(OR=0.59,95%CI=0.38-0.93).In the age stratification analysis,the risk of esophageal cancer in the GG genotype carriers was significantly decreased among individuals>50 years of age(OR=0.59,95%CI=0.36-0.97),but not among individuals≤50 years of age(OR=0.53,95%CI=0.18-1.55).In stratified analysis of smoking,among non-smokers,the risk of esophageal cancer was significantly reduced in patients with GG genotype compared with those with AA genotype(OR=0.36,95%CI=0.18-0.73).However,there was no significant difference between esophageal cancer and the genotypes of rs1927914 in smoking population,smoking status(P>0.05).Attentively,the results of genotyping showed that TLR4 rs11536891 T>C was completely linked to rs7873784 G>C.And there was no significant difference in the distribution of two SNPs genotypes between esophageal cancer patients and normal controls(P>0.05).ConclusionsTLR4 rs1927914 GG genotype significantly reduced the risk of esophageal cancer in Chinese population,while TLR4 rs11536891 T>C,rs7873784 G>C genetic variation did not affect the risk of esophageal cancer.Figure 3;Table 4;Reference 160... |
