Objective To investigate the possible therapeutic effects of curcumin on thyroid papillary carcinoma TPC-1 cells by observing the proliferation,invasion and metastasis of TPC-1 cells,and the changes of Glut1 and MT1-MMP protein expression levels.Methods Human papillary thyroid carcinoma TPC-1 cells in exponential phase were used for experimental study.Experimental group: The control group of curcumin concentration was 0?mol/L,and the experimental group was 10?mol/L,20?mol/L,and40?mol/L for cell intervention.TPC-1 cells were treated with different content of curcumin,using CCK-8 assays to test the inhibitory effect of TPC-1 cell;TPC-1 cells were cultured with curcumin,using transwell assay to evaluate the cell invasion and wound healing assay to detect the migration ability;Real-time PCR and Western blot were used to detect the changes of Glut1 and MT1-MMP m RNA and protein expression in TPC-1 cells by curcumin from the gene and protein levels.Results 1 CCK8 results showed that except the 0?mol/L group,the inhibitory effects of TPC-1 cells at 48 h was significantly higher than that of 24h(P<0.05);2 The invasion experiments results showed that the number of cells passing through the different concentrations of curcumin intervention 0,10,20 and 40?mol/L groups were(196±3.6),(142±4.2),(57±4.5)and(17±5.1)per view.The differences between the groups were statistically significant(P<0.05);3 In the migration experiment,there was a statistically significant difference in the TPC-1 cells between the groups after 24 h and 48 h of drug intervention(P<0.05).The migration of TPC-1 cells was statistically significant at 0h,24 h,and 48 h in the 0?mol/L group(P<0.05).Compared with 0h,there was significant difference in 24 h and 48 h in the 10?mol/L group(P<0.05).While there was no statistical significance in the 20 and 40?mol/L groups(P>0.05);4 Real-time PCR and Western blot showed that the relative expression of Glut1 and MT1-MMP m RNA and protein content were significantly different(P<0.05).Conclusions 1 Curcumin can inhibit the proliferation of thyroid papillary carcinoma TPC-1 cells in a time-and dose-dependent manner.2 curcumin can reduce the invasion and migration ability of thyroid papillary carcinoma TPC-1 cells.3 Curcumin may inhibit the invasion and migration of TPC-1 cells by decreasing the expression of Glut1 and MT1-MMP proteins in thyroid papillary carcinoma TPC-1 cells.Figure6,Table4,Reference 153...
