Objectives Study the effects of Tiaogan Jiedu Recipe and acupuncture on ethology and GFAP expression in brain of epileptic model rats.Methods Take 70 clean SD rats,half male and half female for the experiment.Divide them into 7 groups(10 rats each group)using random number grouping method: blank group,model group,high dose of Chinese medicine group,middle dose of Chinese medicine group,low dose of Chinese medicine group,acupuncture group and sodium valproate group.Establish the epileptic model by chemical ignition method(using PTZ).Judging whether the epileptic model is successful by Lado classification method.Using Tiaogan Jiedu Recipe and Tiaogan Tongdu acupuncture method for intervention therapy.Make ethology test to study the influence on learning,memorizing and emotional ability after the treatment.Using epilepsy seizure latency,seizure grade,Y-maze test and open field test to make the ethology test.Then dissect their brain tissue referring to <Stereotaxic Map of Rat Brain>(Paxinos).The expression of GFAP in brain tissue is determined from dissected brain tissue by immunohistochemical method.Select 10 sections from each group randomly,and observe the same parts of each section using light microscope and calculate the number of positive cells.Results 1.Comparison of normal growth status and body weight: The blank group was normally fed,and they have no epilepsy,with bright hair,no hair loss and normal weight growth.Before treatment,except for the blank group,the other groups of epileptic seizure model rats got weight loss,hair loss,hair gloss deteriorated,loss of appetite and so on.After treatment,body weight of model group was 192.14±8.51,that of high dose of Chinese medicine group was 291.67±18.73,and that of acupuncture group was294.50±16.91.Comparing with the model group,the rats in high dose of Chinese medicine group and acupuncture group add more body weight significantly(P<0.01),and dietary and drinking water increased,and hair gloss of rats in other groups was restored and the depilation was reduced.2.Comparison of epileptic seizure level: There was no epileptic seizure in the blank group during the experiment.After treatment,observing the epileptic seizure grade of rats in each group,statistical significance was found between all groups(P<0.01).The mean rank of model group was 42.29,that of high dose of Chinese medicine group was 11.83,and that of acupuncture group was 12.44.Epileptic seizure grade of high dose of Chinese medicine group and acupuncture group was significantly lower than model group(P<0.01).3.Comparison of epileptic seizure latency: There was no epileptic seizure in the blank group during the experiment.After 21 days of treatment,the latency time of epileptic seizure in model group was 252.71±32.75,in high dose of Chinese medicine group was 646.56±52.88,in acupuncture group was 622.75±47.56,and in sodium valproate group was 629.44±29.02.Comparing with model group,the time of epileptic seizure in high dose of Chinese medicine group,acupuncture group and sodium valproate group was significantly prolonged(P<0.01).4.Comparison of learning and memorizing ability in Y-maze test: Two days of Y-maze avoidance errors were measured after 21 days of treatment.On the first day,Y-maze avoidance errors comparison: avoidance errors of model group were 10.14±1.77,blank group 6.40±1.58,high dose of Chinese medicine group 7.11±1.86,middle dose of Chinese medicine group 8.89±1.27,and acupuncture group 6.88±1.81.Comparing with the blank group,the high dose of Chinese medicine group and the acupuncture group,the avoidance errors in the model group had significantdifference(P< 0.01);compared with the high dose of Chinese medicine group,the avoidance errors in the middle dose of Chinese medicine group had statistical difference(P<0.05).The second day Y-maze avoidance errors count comparison: the blank group avoidance errors count was 5.70±1.49,the high dose of Chinese medicine group was7.00±1.73,the acupuncture group was 6.63±1.69,and the model group was 9.43±1.72.Comparing with the blank group and the acupuncture group,the model group avoidance errors had significant difference(P<0.01);comparing with the model group,the high dose of Chinese medicine avoidance errors had statistical difference(P<0.05).5.Comparison of open field test: After 21 days of treatment,the open-field score of model group was11.14±2.41,that of high dose of Chinese medicine group was 23.33±3.61,that of middle dose of Chinese medicine group was 17.56±2.70,that of acupuncture group was21.88±3.563,and that of sodium valproate group was 19.11±2.93.Comparing with model group,the open-field test score of high dose of Chinese medicine group,acupuncture group and sodium valproate group were significantly higher(P<0.01);and the open-field test score of middle dose of Chinese medicine group was significantly lower than that of the high dose group(P<0.01).6.Comparison of immunohistochemical result: After treatment,the number of GFAP-positive cells in the brain of each group was 24.10±7.08 in blank group,62.71±4.15 in model group,29.00±3.57 in high dose of Chinese medicine group,35.67±3.77 in middle dose of Chinese medicine group,29.25±5.01 in acupuncture group and 34.33±4.66 in sodium valproate group.The number of GFAP-positive cells in blank group,high dose of Chinese medicine group and acupuncture group was significantly lower than that in model group(P<0.01).The expression of GFAP in high dose of Chinese medicine group was significantly lower than that in middle dose of Chinese medicine group(P<0.01).Conclusions1 Pentylenetetrazol chemical ignition can induce epileptic seizure symptoms,increase the expression of GFAP and worsen the performance of ethology tests in rats.2 Both Tiaogan Jiedu Recipe and acupuncture can reduce the level of epileptic seizures and increase the latency of epileptic seizures in epileptic rats,and the high concentration of Tiaogan Jiedu Recipe is obviously better than the middle and low concentration,which has concentration effect.3 Both Tiaogan Jiedu Recipe and acupuncture can improve the ethology performance,improve memorizing and learning ability of epileptic rats,and ameliorate the anxiety and depression of rats,possibly by reducing the expression of GFAP in the
