Objectives To investigate the expression of aquaporins 3(AQP3)and transforming growth factor-?1(TGF-?1)protein and mRNA in serum and decidua tissues of embryo arrest during early pregnancy.Methods 40 patients who were diagnosed as embryo arrest from October 2017 to May2018 in the Affiliated Hospital of North China University of Science and technology were choosed as case group.Another 40 normal pregnants who asked to abortion were used as the control group.We used immunohistochemistry and Western blot to investigate the protein expression of AQP3 and TGF-?1 in decidua tissues.And the expression of AQP3and TGF-?1 in serum we choose ELISA.RT-PCR was used to investigate the mRNA expression of AQP3 and TGF-?1 in decidua tissues.T test and?~2 test were used for data analysis.The consistency analysis used Kappa test.Results 1 The clinical information of the case group and the control inflect that the average age,BMI,the history of abortion history,past history and so on,the difference was not statistically significant,P>0.05.2 The immunohistochemistry's results told that the expression of AQP3 protein in case group was higher than that in control(P<0.05).The expression of TGF-?1 protein in case group was higher than that in control group(P<0.05).3 The relative expression of AQP3 in the case group was 1.11±0.24,which was higher than the control(0.42±0.07).The relative expression of TGF-?1 in the case group was 1.61±0.30,which was higher than the control(0.73±0.13)(P<0.05).4 The results from RT-PCR suggest that the relative expression of AQP3mRNA in the case group was 0.73±0.28,which was higher than control(0.29±0.28).The relative expression of TGF-?1mRNA in the case group was 3.22±1.50 which was higher than control(0.66±0.16)(P<0.05).5 The results from ELISA told that the relative expression of AQP3mRNA in the case group was(3.08±2.67)ng/L,which was higher than control(1.53±2.03)ng/L;The relative expression of TGF-?1mRNA in the case group was(2.43±0.68)ng/L,which was higher than control(1.23±0.18)ng/L,(P<0.05).6 The expression of AQP3 and TGF-?1 in serum and decidua tissue of both groups was related to the occurrence of early embryo arrest(OR>1,P<0.05).7 The expression of AQP3 and TGF-?1 in the serum of the case group were consistent with the levels in the decidua tissues.8 The expression of AQP3 and TGF-?1 in the decidua tissues was consistent with the case group.Conclusions 1 The expression of AQP3 and TGF-?1 in decidua tissues was higher than that of normal pregnancy during early embryo arrest.2 The expression of AQP3 and TGF-?1 in serum was higher than that of normal pregnancy during early embryo arrest.3 The expression of AQP3 and TGF-?1 in the serum was consistent with that in decidua tissues during early embryo arrest,the expression level in serum could represent the expression level in decidua tissue,and they are both related to embryo arrest.4 The expression of AQP3 and TGF-?1 in the decidua tissues was consistent in embryo arrest during early pregnancy.Figure11;Table12;Reference 134...
