Objectives The performances of nonylphenol on ovarian injury in mice were observed,and effects of taurine on nonylphenol-induced ovarian injury in mice were investigated by animal experiments on mice exposed to nonylphenol by taurine.Methods 40 clean-grade female Kunming mice were randomly divided into control group(corn oil + normal saline),exposure group(taurine 0mg/kg + nonylphenol 100mg/kg),lowdose intervention group(taurine 100mg/kg + nonylphenol 100mg/kg)and high dose intervention group(taurine 200mg/kg + nonylphenol 100mg/kg),10 in each group.The control group and the exposed group were intraperitoneally injected with normal saline,and corn oil and 100mg/kg nonylphenol solution were respectively administered;the lowand high-dose intervention groups were intraperitoneally injected with 100mg/kg and 200mg/kg taurine solution,and 100mg/kg nonylphenol solution was administered,5 days a week for 12 consecutive weeks.The general condition of the mice were observed;the weight of the mouse and the wet weight of the ovary were weighed by the electronic balance;the pathological changes of mouse ovarian tissue were observed by HE staining;superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)activity and malondialdehyde(MDA)content in mouse ovarian tissue were detected according to the kit instructions;the expression level of 8-hydroxydeoxyguanosine(8-OHdG)in mouse ovarian tissue was detected by immunohistochemistry;apoptosis of mouse ovarian tissue was detected by TUNEL method.Results 1 The mice in the exposed group had dark hair,less eating,apathetic and poor vitality.The mice in the intervention group recovered with the increase of taurine dose,and the appearance,diet and vitality of the mice were restored.2 The final body weights of the control group,the exposed group and the low-and high-dose intervention groups were(38.58±1.92)g,(35.97±1.71)g,(37.11±1.99)g and(38.45±1.97)g,respectively,and the difference was statistically significant(P<0.05),the exposure group was lower than the control group,and the high dose intervention group was higher than the exposure group.3 The wet weight of ovary in the four groups of mice was(0.29±0.04)g,(0.22±0.04)g,(0.24±0.06)g and(0.26±0.03)g,respectively,and the difference was statistically significant(P<0.05),the exposure group was significantly lower than the control group.The highdose intervention group was higher than the exposure group.4 The total number of follicles in the ovary of the mice in the exposed group decreased,vacuolar changes,follicles in the atresia increased,inflammatory cell in the interstitial was infiltrated;ovarian tissue morphology in the intervention group was restored,and the high-dose intervention group was the most obvious.5 The SOD activity of the ovarian tissues of the four groups of mice were(53.39±4.70)U/mgprot,(26.07±6.54)U/mgprot,(28.59±3.80)U/mgprot and(41.13±3.58)U/mgprot,the GSH-Px activity were(7.58±1.35)U/mgprot,(1.91±0.93)U/mgprot,(3.12±1.16)U/mgprot and(4.62±0.62)U/mgprot,the MDA content were(2.28±0.41)nmol/mgprot,(3.72±0.38)nmol/mgprot,(3.25±0.20)nmol/mgprot and(2.87±0.37)nmol/mgprot,the difference was statistically significant(P<0.05),compared with the exposed group,the activity of SOD and GSH-Px in the ovarian tissue of the high-dose intervention group was significantly increased,and the MDA content was significantly decreased.6 The mean optical density values of 8-OHdG expression in ovarian tissue of the four groups of mice were 0.0117±0.0061,0.0751±0.0099,0.0647±0.0066 and 0.0446±0.0087,respectively,the difference was statistically significant(P<0.05),and the exposure group was significantly higher than that in the control group,the high-dose intervention group was significantly lower than the exposure group.7 The apoptosis rates of ovarian tissue positive cells in the four groups of mice were 5.60%,44.25%,32.95% and 26.00%,respectively,the difference was statistically significant(P<0.05),and the exposure group was significantly higher,in the control group,the intervention group was significantly lower than the exposure group.Conclusions 1 Taurine has a protective effect on the general condition and body weight of mice induced by nonylphenol;2 Taurine has an effect on the morphological damage of mouse ovarian tissue induced by nonylphenol.With the increase of taurine dose,the effect is more obvious;3 Taurine has an antagonistic effect on oxidative damage of mouse ovarian tissue induced by nonylphenol;4 Taurine has a protective effect on the apoptosis of mouse ovarian tissue induced by nonylphenol,and the effect of high dose of taurine is more obvious.Figure 9;Table 5;Reference 151...
