A Preliminary Study On Serum MiRNA Screening And Its Diagnostic Value Of OSAHS

?Objective?To observe the differentially expressed miRNAs in OSAHS patients without complications and OSAHS patients with hypertension,with an attempt to explore its diagnostic value for OSAHS.? Methods ? The subjects included patients who visited the Department of Otorhinolaryngology,the Union Hospital of Tongji Medical College,Huazhong University of Science and Technology and healthy controls who recruited from the Health Screening Center of the same hospital.All participants underwent physical checkups,daytime sleepiness assessment,overnight polysomnography(PSG)and tests of blood biochemistry.The serum miRNA profiles of OSAHS patients without complications,OSAHS patients with hypertension and healthy controls were assessed by microarray technology,and then differentially expressed miRNAs were verified by quantitative real-time PCR(qRT-PCR).Bioinformatics assessment of validated miRNAs to further determine their functions and signaling pathways,and preliminarily explore their diagnostic value.? Results ?(1)The microarray exhibited that compared to the healthy controls,12 differentially expressed miRNAs were found in OSAHS patients without complications,and 33 were found in OSAHS patients with hypertension.(2)MiR-126-3p,let-7d-5p,miR-320 b,miR-145-5p were verified to be down-regulated in OSAHS patients without complications and miR-126-3p,miR-107,miR-26a-5p were validated to be down-regulated in OSAHS patients with hypertension.(3)Bioinformatics analysis predicted that the potential target genes of these miRNAs might be involved in metabolism,and the regulation of endothelial cells and nervous system.(4)The receiver operating characteristic(ROC)analysis showed that the areas under the curve(AUC)for miR-126-3p?let-7d-5p?miR-145-5p and miR-320 b are 0.822(95% confidence interval(CI):0.671~0.973),0.778(95%CI:0.598~0.958),0.849(95%CI:0.711~0.986),0.751(95%CI:0.570~0.932)in identify OSAHS without complications,the AUC for miR-126-3p,miR-26a-5p and miR-107 are 0.871(95%CI:0.747~0.995),0.729(95%CI:0.543-0.914),0.733(95%CI:0.552-0.915)in identify OSAHS patients with hypertension.?Conclusion?By using the miRNA microarray technology,a cluster of miRNAs related to OSAHS has been found,which might be used as early warning and diagnostic markers of OSAHS.
