Objective To investigate the relationship between serum thioredoxin 2(Trx2)levels and white matter hyperintensity and insomnia.Methods From November 2017 to June 2018,164 patients with non-specific neurological symptoms such as dizziness were enrolled in the Affiliated Hospital of Shihezi University Medical College.All patients underwent a whole brain scan using the DISCOVERY MR750 3.0T high-field magnetic resonance imager.Two neurological physicians used the Fazekas scale to evaluate the white matter lesions in patients,and based on ICSD-3 to diagnose insomnia.According to the results of the examination,the study subjects were divided into simple insomnia patients(simple insomnia group,42 cases),simple white matter high signal patients(simple white matter hyperintensity group,40 cases)and white matter high signal with insomnia patients(white matter hyperintensity with insomnia group,82 cases).In addition,45 patients who underwent physical examination in our hospital were selected as the control group.We used PSQI,ESS,GSES,FIRST,FFS to evaluate the insomnia of all patients and examiners.Serum Trx2 levels were detected by ELISA.Results(1)There was no significant difference in gender composition,age,education level,smoking,drinking,and homocysteine between the control group,the simple insomnia group,the simple WMH group,and the WMH with insomnia group(P>0.05).There was a statistically difference in diabetes and hypertension between the groups(P<0.05).(2)There was no significant difference in ESS between the groups(P>0.05).The FFS,GSES,FIRST and PSQI of the WMH with insomnia group were higher than those of the insomnia group and the control group,the difference was statistically significant(P< 0.001);FFS,GSES,FIRST and PSQI in the insomnia group were higher than those in the control group,and the difference was statistically significant(P<0.001).There was no significant difference in FFS,GSES,FIRST and PSQI between the simple WMH group and the control group(P>0.05).(3)The serum Trx2 level in the simple insomnia group was higher than that in the control group(P<0.001).The serum Trx2 level in the simple WMH group was higher than that in the control group(P<0.001).There was no significant difference of serum Trx2 level between simple WMH group and simple insomnia group(P>0.05).The serum Trx2 level in WMH with insomnia group was higher than simple WMH group,simple insomnia group and control group and the difference was statistically significant(P < 0.001).(4)Pearson or Spearman correlation analysis showed that serum Trx2 level was positively correlated with Fazekas score,and the results were statistically significant(P < 0.001).Serum Trx2 levels,Fazekas scores were positively correlated with PSQI scores,and the results were statistically significant(P < 0.001).Conclusion(1)Serum Trx2 level is elevated in patients with white matter hyperintensity or insomnia,and is significantly elevated in patients with white matter hyperintensity and insomnia,which may provide new biological indicators for white matter hyperintensity and insomnia.(2)Serum Trx2 level increases with the increase of white matter hyperintensity,and the severity of white matter hyperintensity can be assessed by measuring serum Trx2 level.
