BACKGROUND:Osteoarthritis,as a disease that seriously afflicts the quality of life of middle-aged and elderly people,has a wide variety of pathogenic factors.Matrix metalloproteinase-3(MMP-3)and galectin-3(Galectin-3)play an important role in the pathological process of osteoarthritis.There may be some potential relationship between them.We have consulted a lot of literature and materials,but have not seen any relevant reports.OBJECTIVE:To detect the expression levels of MMP-3 and Galectin-3 in the synovial tissue of osteoarthritis,and to investigate the relationship between MMP-3 and Galectin-3 in the pathogenesis of osteoarthritis.METHODS:Synovial tissues were obtained from the 60 patients undergoing total knee arthroplasty or knee arthroscopic diagnosis and treatment surgery and 20 healthy patients suffering from amputation caused by trauma or meniscus or knee ligament injury caused by acute injury.According to the WOMAC Osteoarthritis Index,the OA observation group was divided into three groups:mild,moderate,and severe,with 20 cases in each group.Synovial membrane of the knee joint was collected during the operation in both groups and stored in 4%paraformaldehyde for future use.Immunohistochemical method was used to detect the expression of MMP-3 and Galectin-3 in synovial tissue of osteoarthritis patients,and continuous correction chi-square test and Pearson correlation analysis were performed by statistical software SPSS 22.RESULTS:Immunohistochemistry results showed that MMP-3 was positive in 2(10%)in the non-osteoarthritis group,positive in 7(35%)samples in the mild-osteoarthritis group,positive in 13(65%)samples in the medium-osteoarthritis group and positive in 18(90%)samples in the severe-osteoarthritis group.Galectin-3 was positive in 1(5%)samples in the non-osteoarthritis group,positive in 6(30%)samples in the mild-osteoarthritis group,positive in 12(60%)samples in the medium-osteoarthritis group and positive in the in 17(85%)samples in the severe-osteoarthritis group.The positive expression rate of MMP-3 and Galectin-3 in osteoarthritis group was significantly higher than that of non-osteoarthritis group(?~2=15.000/15.152,P<0.001)in continuous correction.Pearson correlation analysis showed a significant positive correlation between the expression of synovial MMP-3 and Galectin-3 in patients with osteoarthritis and the WOMAC index(r=0.924/0.940,P<0.001),There was a significant positive correlation between the expression of MMP-3 and Galectin-3 and the rate of synovitis(r=0.922/0.925,P<0.001),and the expression of MMP-3 and Galectin-3 was also significantly positive(r=0.926,P<0.001).CONCLUSION:1.The expression of MMP-3 and Galectin-3 in synovial membrane of OA patients is significantly higher than that of non OA patients,which can help early diagnosis of OA.2.The expression of MMP-3 and Galectin-3 in synovial specimens of OA patients was significantly positively correlated,and both of them played a synergistic role in OA.
