Objective1.Explore the role of long-chain non-coding RNA LINC00052 in the development of CRC.2.Identify the mechanism of LINC00052 in CRC.Methods1.qRT-PCR was used to detect the expression of LINC00052 in 24 pairs of colorectal cancer tissues and adjacent tissues.2.The LINC00052 overexpression plasmid was constructed and its interference fragment was synthesised.The colony formation assay,MTS assay,transwell chamber assay and wound healing assay were performed to detect the biological function of LINC00052 in HT-29 and SW620 cells.3.Predict microRNAs that interact with LINC00052 by using bioinformatics software.A dual luciferase reporter assay was performed to verify the interaction between LINC00052 and microRNA.4.qRT-PCR was used to detect the expression of microRNA in CRC tissues,microRNA mimics and inhibitors were purchased for detecting the biological functions of microRNA.5.Bioinformatics website was used to predict downstream target genes of microRNA,a dual luciferase assay was used to detect the relationship between microRNA and target genes.6.The expression of target gene in colorectal cancer tissues was detected by qRT-PCR and WB,and the effects of microRNA and LINC00052 on the migration and invasion of CRC cells were detected by transwell and wound healing assays.7.Nude mouse metastasis model of LINC00052 overexpression was performed to detect the effect of LINC00052 on colorectal cancer metastasis in vivo.Results1.LINC00052 is down-regulated in CRC tissues and can inhibit the proliferation and metastasis of CRC cells.2.LINC00052 can down-regulate the expression of miR-574-5p,and miR-574-5p is up-regulated in colorectal cancer tissues,which can promote the proliferation and metastasis of colorectal cancer cells.3.LINC00052 can up-regulate CALCOCO1 expression in combination with miR-574-5p,thereby inhibiting the metastasis of colorectal cancer cells.4.CALCOCO1 is down-regulated in colorectal cancer tissues,and miR-574-5p can rescue the inhibiting of LINC00052 on colorectal cancer cell metastasis.5.Overexpression of LINC00052 in animal experiments can inhibit the metastasis of colorectal cancer cells in vivo.ConclusionLINC00052 inhibits colorectal cancer metastasis by competitively binding miR-574-5p to up-regulate CALCOCO1 expression.
