Objective:To explore protective effect and possible mechanism of quercetin on the overload pressure rat.Methods:The rats which pressure overload was constructed by abdominal aorta stenosis.40 SD rats were divided into Sham group(n=10);AAC group(n=10),Quercetin group(n=10),EX527 group(n=10).Sham group and AAC group were given daily saline and intraperitoneal injection;Quercetin group was given Quercetin 50mg/kg.d daily for intragastric administration and normal saline intraperitoneal injection;EX527 group was given SIRT1 specific inhibitor EX527 5mg/day via intraperitoneal injecting and quercetin 50mg/kg.d for intragastric administration.Evaluating the cardiac function,the thickness of the left ventricular posterior wall;the degree of myocardial fibrosis and the expressions of ColI,Col?,SIRT1 and NF-kB.Results:Quercetin could improve the thickness of posterior wall of left ventricle(sham:1.587±0.136AAC:2.657±0.355,Quercetin:1.800±0.200,EX527:2.700±0.306,P=0.001,F=37.304),attenuating myocardial fibrosis(sham:8.515±1.343/AAC:23.832±1.095/Quercetin:13.260±0.674/EX527:24.162±1.312/F=278.741/P=0.000)and decreasing the expression of ColI(sham:1.000±0.000,AA:3.132±0.372/Quercetin:1.556±0.164/EX527:2.819±0.368,F=82.083,p=0.000)and Col?(sham:1.000±0.000/AAC:2.395±0.437/Quercetin:1.583±0.287/EX527:2.434±0.461,F=23.608/P=0.024)in rats after the surgery of abdominal aorta constriction.And enhancing SIRT1 expression[sham:1.000±0.000,AAC:0.364±0.071,Quercetin:1.138±0.070,EX527:0.293±0.092/F=240.539/P=0.000j.EX527 increasing the expression of Coll,Col? and NF-kB in rats after the therapy of quercetin(P=0.001,0.024,0.023).And decreasing the expression of SIRT1(P=0.000).Conclusion:Quercetin can decrease the extent of myocardial fibrosis and alleviate left ventricular hypertrophy in rats after the surgery of abdominal aorta constriction and the mechanism may be related to the SIRT1/NF-kB pathway.
