Objective:To study the expression of Sirtuin 3(Sirt3)in trophoblast cells at different pregnancy status(Normal Control/Normotensive pregnancies,NC and Pre-eclampsia,PE),and to detect the effect of reduced expression of Sirt3 in the development of pre-eclampsia(PE).Methods:From September 2016 to March 2017,12 PE placental tissue specimens and 12 normal cases were acquired in obstetrical department of the First Affiliated Hospital of Chongqing Medical University.And all delivery methods are cesarean section.The expression of Sirt3 was determined in the placenta from normal case and PE group by Immunofluorescence technique(IF)and Western blot(WB).Sirt3 was knockdown in HTR8/SVneo cell line through lentiviral transfection technique.WB was used to identify the expression of Sirt3 in Sirt3-KD HTR8/SVneo cell after lentiviral transfection.The experiments were composed of Vehicle group and LV-Sirt3 group,then the production of ATP was measured through ATP Kit to estimate the function of mitochondrial productivity.And apoptosis was determined by FCM and CCK-8.Finally,active caspase-3?IL-1?and MMP2 were evaluated by WB.Results:Sirt3 was less expressed in the PE group comapared with NC group.The production of ATP was reduced in LV-Sirt3 group,which shows that the dysfunction of mitochondria in energy generation.Meanwhile,apoptosis and inflammatory factor were significantly increased in LV-Sirt3 HTR8/SVneo cell in comparison with Vehicle group.Besides,aggression-ralated factor MMP2 was decreased in LV-Sirt3 group.Conclusion:The downregulation of Sirt3 results in the apoptosis of trophoblast,which may be associated with the pathogeny of PE.
