Objective With the extensive use of proton pump inhibitors(PPI),the adverse reactions it may cause have been gradually taken seriously. The objective of this article is to systematically research the relationship between PPI and the risk of hypomagnesemia and providing evidences for clinical medication. Methods A systematic search about the relationship between the use of PPI and hypomagnesemia was performed on database including Pubmed,Embase,Cochrane,CNKI,Wanfang and VIP.After the literatures were screened one by one,the relevant data were extracted and analyzed by Revman 5.3 software. The heterogeneity was tested by Q test and I2 test.Sensitivity analysis was performed by eliminating studies one by one. Results 12 studies including 17,039 patients were analyzed.Heterogeneity test showed mild heterogeneity in this study(P=0.18,I2=27%).Meta analysis showed that the occurance of hypomagnesemia in patients taking PPI was significantly higher than those who did not take PPI(OR=2.20,95%CI:1.78-2.72,P<0.00001).Further subgroup analysis based on the differences in criteria for hypomagnesemiaand patient types came the same conclusion that the incidence of hypomagnesemia in PPI users was higher than that in non-PPI users.Sensitivity analysis showed that there was no significant change in the combined effect after any study was excluded. Conclusions PPI may contribute to the occurance of hypomagnesemia, suggesting that more attention should be paid to the serum magnesium levels in patients who have been taking PPI for a long time. However, most of the studies included in this paper are not randomized controlled trials, which may result in potential bias, such as selection bias.More clinical studies with higher quality of evidence are needed in the future.
