The Effects Of RNF10 On The Function Of Vascular Smooth Muscle Cells Induced Exposed To High Glucose | | Posted on:2020-09-14 | Degree:Master | Type:Thesis | | Country:China | Candidate:S Y Li | Full Text:PDF | | GTID:2404330590480059 | Subject:Internal Medicine | | Abstract/Summary: | PDF Full Text Request | | OBJECTIVES:To investigate the protective effects of RNF10 on vascular smooth muscle cells(VSMCs)exposed to high glucose.METHODS:Vascular smooth muscle cells(VSMCs)were induced by high glucose(4.5g/L)to proliferate excessively and deposit extracellular matrix in vitro.According to the culture medium and adenovirus transfection,the cell experiment was divided into four groups:Control group;High glucose group(HG group);High glucose+Adenovirus Green fluorescent protein group(HG+Ad-GFP group);High glucose+overexpression RNF10 group(HG+Ad-RNF10 group).Qualitative analysis of VSMCs adenovirus transfection rate was observedunder fluorescence microscope;Both CCK-8 and EdU assays were used to detect VSMCs proliferative rate;Flow cytometry verified cell cycle distribution of VSMCs;Western blot analyzed the expressions of RNF10、CyclinD1and CDK4 protein in VSMCs;qPCR analyzed the expressions of MMP-2and TIMP-2 mRNA in VSMCs.Results:(1)RNF10 significantly inhibited the proliferation of VSMCs exposed to high glucose(P<0.05);(2)RNF10 significantly inhibited the transition from G0-G1 to S phase of VSMCs exposed to high glucose(P<0.05);(3)RNF10 significantly inhibited the expressions of Cyclin D1 and CDK 4 in VSMCs exposed to high glucose(P<0.05);(4)RNF10 significantly inhibited the transcription levels of MMP-2 and TIMP-2(both P<0.05),however the inhibition of TIMP-2 was more obvious.Eventually RNF10 significantly inhibited of MMP-2/TIMP-2(P<0.05).CONCLUSION:RNF10 can significantly improve the function of VSMCs exposed to high glucose,which may be related to the inhibition of VSMCs proliferation and extracellular matrix accumulation. | | Keywords/Search Tags: | RNF10, vascular smooth muscle cells, proliferation, extracellular matrix | PDF Full Text Request | Related items |
