Objective: To investigate the feasibility and effectiveness of PPCNg-mediated human amniotic mesenchymal stem cells(hAMSCs)in the treatment of intrauterine adhesions(IUA).Methods: hAMSCs were cultured in different concentrations of PPCNg.After 48 hours,the cell morphology was observed with the fluorescence microscope,and the cytotoxicity of the material was measured by CCK-8.Thirty female SD Rats were randomly divided into sham group,model group,PPCNg group,hAMSCs group and PPCNg+hAMSCs group.On the second week after modeling,sham group was not treated,while the other four groups were injected with PBS,PPCN-g,PBS suspension containing hAMSCs and PPCNg suspension containing hAMSCs in the uterine cavity respectively.On the second week after the injection,the histological observation was performed to detect the number of endometrial glands and the ratio of fibrosis area.Immunohistochemistry was performed to detect the expression of TGF-?1,VEGF,VIM,?-SMA,CK19,E-Ca in the endometrium.The red fluorescence distribution was observed under fluorescence confocal microscope.Results: In any concentrations of PPCNg,hAMSCs grew well by adherence and PPCNg was not cytotoxic.Compared with sham group,the gland number in model group decreased(P=0.000)and the fibrosis area ratio increased(P=0.000).Compared with model group,the gland number of in PPCNg group,hAMSCs group and PPCNg+hAMSCs group increased(P=0.000),and the fibrosis area ratio decreased.Compared with model group,the expression of TGF-?1,VEGF,VIM and ?-SMA in PPCNg+hAMSCs group decreased significantly(P=0.000),the expression of CK19 and E-Ca in PPCNg+hAMSCs group increased significantly(P=0.000).PKH26-labeled hAMSCs in PPCNg+hAMSCs group were much more than those in hAMSCs group.Conclusion: PPCNg has almost no cytotoxicity and can significantly improve the therapeutic effect of hAMSCs in IUA.
