Interaction Between Genes Of CREB1-BDNF-NTRK2 Pathway Is Possibly Associated With Suicide Attempt In Young People

Objective To explore the interaction between cAMP response element binding protein I(CREB1),brain-derived neurotrophic factor(BDNF)gene and Neurotrophic Tyrosine Kinase Receptor Type 2(NTRK2)gene relevant for young people suicide attempt.Methods One single nucleotide polymorphism(SNP)of CREB1 gene(rs7569963?rs4675690),BDNF gene(rs1491851)and NTRK2(rs1147198?rs11140800?rs1565445)were tested by IMLDRtm multiplex single nucleotide polymorphism typing in 70 suicide attempt completers and 111 healthy controls.Analyse gene-gene interaction by generalized multifactor dimensionality reduction(GMDR).Results No significant differences were found between the case group and control group at 6 SNPs(rs7569963,rs4675690,rs1491851,rs1147198,rs1140800,rs1565445)in genotype frequencies or allele frequencies(p>0.05).GMDR analysis showed that there was no interaction between CREB1 gene and NTRK2 gene,CREB1 gene and BDNF gene or CREB1-BDNF-NTRK2,but indicated the interactions between BDNF gene and NTRK2 gene which included a two-locus combination(rs1491851 ? rs1147198),a three-locus combination(rs1491851 ? rs1147198 ? rs11140800)and four-locus combination(rs1491851 ? rs1147198 ? rs11140800 ? rs1565445).The four-locus combination(Bal.Acc.CV testing 0.6325,CVC consistency 10/10,permutation test p value 0.032-0.033)was the best model.Conclusion There are interactions between BDNF gene and NTRK2 gene,which may be associated with suicide attempt in young people.But no evidence proved that there was interaction between CREB1 gene and NTRK2 gene,CREB1 gene and BDNF gene or CREB1-BDNF-NTRK2.
