Objective: 1.To construct CD133-targeted multifunctional fluorescent/ultrasonic nanoparticles and observe it's general properties.2.To disscuss the in vitro targeting ability of CD133 NPs.3.To disscuss the ability on ultrasound / fluorescent imaging for CD133 positive pancreatic cancer stem cell of CD133 NPs.4.To disscuss the efficacy of CD133 NPs synergism of HIFU treatment for nude mouse pancreatic cancer xenograft.Methods: 1.To construct CD133-targeted multifunctional fluorescent/ultrasonic nanoparticles and Ig G-linked non-targeted fluorescent/ultrasonic nanoparticles by ATRP methods,characterize to determines it appearance and size.2.Proliferation inhibition experiment and apoptosis experiment.3.Flow cytometry was used to detect the CD133 expression ratio of three tyes of pancreatic cancer cell line.The cells were incubated with CD133 NPs or Ig GNPs to observe the combination in vitro.4.Ultrasound imaging and small animal fluorescence imaging were performed at different time after micelle was injected.5.HIFU irradiate polyacrylamide gel to compare the changes of ultrasonic change before and after irradiation in ablation area,and the coagulation necrosis volume was calculated.The tumor of HIFU irradiated after tail vein injection at different time was observed by ultrasonography,and pathological analysis was performed.Results: 1.The mean diameter of micelle was approximately 100 nm.2.The combination rate of CD133 NPs with CD133 highly expressed Bx PC-3 cells was higher than that of the other two pancreatic cancer cells line.3.The ultrasonography of tumor 24 h after CD133 NPs intravenous injection showed that the echo enhancement was stronger than that of Ig GNPs at tumor site,and the fluorescent intensity was the strongest in the tumor site 24 hours after CD133 NPs intravenous injection.4.The ablation area of the tumor was larger 24 hours after the injection of CD133 NPs than that of Ig GNPs,p<0.05.Conclusion: CD133 NPs target CD133 positive pancreatic cancer stem cells specifically,which can be detected by ultrasound/fluorescence imaging,and can enhance the ablation effect of HIFU on pancreatic cancer in nude mice.
