OBJECTIVE-Dermatopontin(DPT)is a member of non-collagen proteins and involves in regulating interactions between other components of extracellular matrix.High temperature requirement A1(HtrA1)serine protease can be induced to secrete under stress,degradating some abnormal components of extracellular matrix in physiological or pathological process.We found DPT and HtrA1 had a high level of expression in white adipose tissue,but their functions have been unclear.This study was to observe the gene expression of DPT and HtrA1 in adipose tissue remodeling of different metabolic states.METHODS-Searching public databases to observe the gene expression profiles of DPT and HtrA1,and real-time quantitative PCR and western blot were used to verify the gene expression profiles in various tissues of mouse;Detecting the expression of DPT and HtrA1 in Vascular stromal cells and adipocytes resided in adipose tissue,and also during 3T3-L1 preadipocyte differentiation process;Comparing expression of DPT in adipose tissue of normal and obese mouse,and HtrA1 in adipose tissue of normal and obese and short time high fat diet feeding mouse;Observing the expression of DPT and HtrA1 by treating certain concentrations of rosiglitazone and cobalt chloride with 3T3-L1 differentiated adipocytes.RESULTS-1)DPT expressed high in white adipose tissue;DPT was mainly synthesized and secreted from adipocytes;High fat diet induced DPT expression in white adipose tissue obviously;Rosiglitazone could down-regulate DPT expression in 3T3-L1 adipocytes.2)HtrA1 expressed high in adipose tissue;HtrA1 was mainly synthesized and secreted from adipocytes;Short time high fat diet induced HtrA1 expression in adipose tissue obviously;Hypoxia could up-regulate HtrA1 expression in 3T3-L1 adipocytes.CONCLUSIONS-Our results described the obvious differences of DPT and HtrA1 expression in adipose tissue of different metabolic states,it was suggested they were likely to participate in the development of obesity,but the mechanisms have been unclear.
