Stroke is one of the most deleterious diseases with high morbidity and mortality.The function loss of brain vessels is one of the reasons why stroke is hard to cure,therefore improving the function of blood vessel in ischemic area is an important strategy to treat stroke.Dl-3-N-butylphthalide(NBP)is a small molecule drug that is independently developed in China.It was shown to increase the blood flow in ischemic area and protect the brain.NBP has many mechanisms including inhibiting apoptosis,promoting neurogenesis and angiogenesis.But at present there are few studies that focus on the effect of NBP on the cerebral blood vessels.Especially,researches that combine cerebral vascular imaging with molecular biology are lacking.In the present study,we combined synchrotron radiation angiography(SRA)method with molecular biology assessment of key gene expressions to study the effect of NBP on the functional recovery and blood vessel function after stroke.The main conclusions of the study are summarized in four parts.1.The stroke rats received continuous NBP treatment showed less body weight loss,significantly improved neurological function evaluated by mNSS after ischemia.The brain infarct volume assessed 1 week after stroke and brain atrophy volume assessed 3 week after stroke were both decreased in the rats with continuous NBP treatment.These two results indicated the protection effect of NBP in ischemic rats.2.The gene expression level of key factors in the thrombosis system and vasoconstriction system were in chaos after ischemic stroke,and NBP treatment regulated most of the gene expressions to their normal level.3.Thrombosis was observed by SRA in rats received tMCAO surgery.Rats treated with NBP had reduced incidences of thrombosis,which proved the thrombosis inhibition effect of NBP in ischemic arteries.4.In the acute and subacute phase of stroke,NBP showed a vasodilation effect in a few hours after NBP treatment.The results showed that NBP ameliorated the vasoconstriction after the ischemic/reperfusion in the acute phase,and that NBP had vasodilation effect in the subacute phase on the vessels in ischemic core.The results in the present study support that NBP improved the function of cerebral vessels and is beneficial for stroke recovery.
