| Backgroud:Hypertension is a clinical syndrome characterized by elevated blood pressure in the systemic circulation of the arteries and is a major public health problem worldwide.And endothelial dysfunction is an important physiological and pathological basis of hypertension.Acupuncture is an effective means to regulate blood pressure and improve various symptoms caused by elevated blood pressure.Studies have shown that high blood pressure from Yangming theory is effective and acupuncture at Yangming meridian points has the effect of promoting gastrointestinal secretion of Ghrelin,while Ghrelin regulates The PI3K/AKT/e NOS signaling pathway is the key pathway for endothelial dysfunction.Based on this,this experiment begins with the PI3K/AKT/e NOS signaling pathway by observing the spontaneous hypertensive rat model,and observes the electroacupuncture at Yangming meridian.The effects of blood pressure,endothelial function,Ghrelin and PI3K/AKT/e NOS signal transduction pathways in spontaneously hypertensive rats provide new ideas for acupuncture theory and clinical prevention and treatment of hypertensionObjective:Spontaneously hypertensive rats were used as research objects.The effects of electro-acupuncture at Yangming Meridian on blood pressure and endothelium protection in spontaneously hypertensive rats were studied by tail-stem method,histomorphology,enzyme-linked assay and Western-blot.The effects of Ghrelin and PI3K/AKT/e NOS signal transduction pathways clarify whether hypertension from Yangming treatment activates Ghrelin-mediated PI3K/AKT/e NOS signaling pathway,protects endothelial function,and exerts anti-hypertensive effects.Methods:Twenty SPF-level spontaneously hypertensive rats(SHR)and normal blood pressure rats(WKY)were selected,male,8 weeks old,weighing 250-300 g,free to eat and drink.Ten WKY rats were in the normal blood pressure group.Twenty SHR rats were randomly divided into spontaneous hypertension group and electro acupuncture Yangming group according to random number table method.The selection of acupoints in the Yangming group of electro acupuncture and the points of Quchi,Zusanli,and Fenglong.Rats in each group were measured for blood pressure in rats at different time points by tail sleeve method.In addition to the spontaneous hypertension group and the normalblood pressure control group,the electroacupuncture Yangming group received 5 electro acupuncture treatments per week for 4 weeks.Spontaneous hypertension group and normal blood pressure control group: daily binding for 20 min,the same time as acupuncture group,NO treatment,5times a week for 4 weeks.After 30 days,the rats were sacrificed and one side of the carotid artery was taken to observe the pathological changes of blood vessels.Determination of Ghrelin,NO,ET-1 and TXA2 in rat plasma by ELISA.Western blotting of protein expression of PI3 K,AKT and e NOS in aorta.Results:part 1 Effect of electroacupuncture at Yangming Meridian on blood pressure1.Electroacupuncture at Yangming Meridian can reduce blood pressure in hypertensive rats : After 4 weeks of acupuncture intervention,the SBP of the electroacupuncture Yangming group was 10.65±4.28 mm Hg lower than that before acupuncture,the difference was statistically significant(p<0.01);DBP decreased by2.33±4.87 mm Hg before acupuncture.,the difference was statistically significant(p<0.05).2.The electroacupuncture at Yangming meridian point has an immediate effect,and the immediate effect in this experiment is better within 30 min.This may be related to the sensitivity of blood pressure to acupuncture:The results of paired samples t-test after SBP and DBP acupuncture for 30 min,1h and 2h showed that the SBP and DBP of acupuncture for 30 min were lower than that of acupuncture for 1h and 2h,the difference was statistically significant(p<0.05);There was NO significant difference in SBP between acupuncture at 30 min and acupuncture at 1 h for 2 weeks(p>0.05).After 4 weeks of treatment and 2 weeks of treatment,the antihypertensive effect of acupuncture within 30 minutes is better.3.Compared with acupuncture at 2 weeks and 4 weeks,the blood pressure decreased by a different degree;the antihypertensive effect of the fourth week of acupuncture treatment was better than that of the second week,which may be related to the persistent effect of acupuncture:By comparison,the electroacupuncture group Yangming group had a decrease of 11.88±1.15 mm Hg(p<0.05)at 2 weeks after acupuncture,and the difference was statistically significant.The DBP difference was not truly significant;Compared with before acupuncture,the SBP decreased by 16.2±0.54 mm Hg(p<0.05),and the difference was significant.The DBP decreased by 22.25±2.63 mm Hg(p<0.05).The difference was statistically significant.SBP and DBP were lower than SBP and DBP before acupuncture for 4weeks.SBP and DBP after 4 weeks of acupuncture were lower than SBP and DBP after 2 weeks of acupuncture.The difference is also statistically significant.Compared with 2 weeks and 4 weeks,the blood pressure decreased by acupuncture.The anti-hypertensive effect after 4 weeks of acupuncture was better than that of acupuncture at 2 weeks.Part 2 Effect of Electroacupuncture at Yangming Meridian on Vascular Endothelial Function and Tube Wall Morphology1.Electroacupuncture at Yangming Meridian can increase plasma NO concentration in SHR rats and decrease plasma ET-1 and TXA2 levels in SHR rats.:Compared with the normal blood pressure group,the plasma concentrations of ET-1 and TXA2 in the spontaneous hypertension group were significantly increased (P<0.01).Compared with the spontaneously hypertensive rats,the electroacupuncture was positive.The plasma concentrations of ET-1 and TXA2 in the Yangming group were significantly lower(P<0.05).2.Electroacupuncture at Yangming Meridian can improve the morphology of vascular endothelium: After 4 weeks of acupuncture treatment,the electroacupuncture of Yangmingjing rats decreased the number of vascular endothelial cells and reduced the damage of the vessel wall.From the morphological point of view,acupuncture has the effect of protecting the vascular endothelium.Part 3 Effect of Electroacupuncture at Yangming Meridian on PI3K/AKT/e NOS Pathway and Ghrelin1.Electroacupuncture at Yangming Meridian can activate and up-regulate the signal expression of PI3K/AKT/e NOS:(1)Compared with normal blood pressure rats,the expression of e NOS protein in the abdominal aorta of rats with spontaneous hypertension decreased,the difference was extremely significant(P<0.01);Compared with spontaneously hypertensive rats,the expression of e NOS protein in the abdominal aorta of rats in the electro-acupuncture Yangming group was significantly increased(P<0.05).(2)Compared with the normal blood pressure group,the expression of AKT protein in the abdominal aorta of the electro-acupuncture-yangming group was significantly increased(P<0.05).(3)Compared with the normal blood pressure group,the expression of PI3 K protein in the abdominal aorta of the spontaneous hypertension group was significantly lower(P<0.01).Compared with the spontaneously hypertensive rats,the expression of PI3 K protein in the abdominal aorta of the electroacupuncture Yangming group was significantly increased(P<0.05).2.Electroacupuncture at Yangming Meridian Acupoints can Increase the Content of Ghrelin in Plasma of SHR Rats:Compared with the normal blood pressure group,the concentration of Ghrelin in the plasma of spontaneously hypertensive rats decreased,and the difference was significant(P<0.05).Compared with spontaneously hypertensive rats,the plasma concentration of Ghrelin in the electro acupuncture yangming group was significantly higher(P<0.05).Conclusion:1.Electroacupuncture at Yangming Meridian can reduce blood pressure in hypertensive rats2.Compared with acupuncture at 2 weeks and 4 weeks,the blood pressure decreased by a different degree;the anti-hypertensive effect of the fourth week of acupuncture treatment was better than that is better within 30 min.This may be related to the sensitivity of blood pressure to acupuncture4.The concentration of ET-1 and TXA2 in the plasma of rats with electro-acupuncture Yangming group decreased,and the concentration of NO increased.Acupuncture can activate regulate the concentration of NO,and increase the secretion and release.It inhibits the damage of cytotoxic substances to the endothelium and increases the vasodilation function,thereby lowering blood pressure.and After 4 weeks of acupuncture treatment,the number of vascular endothelial cells was decreased,and the vessel wall was reduced.The degree of damage is reduced,and the effect of acupuncture on protecting the vascular endothelium is illustrated from the morphological point of view.5.Electro-acupuncture at Yangming Meridian can up-regulate the signal of PI3K/AKT/e NOS to pathway and increase,increase the concentration of ghrelin in the plasma of SHR rats,protect the vascular endothelium,and exert the anti-hypertensive effect.In summary,the blood pressure of hypertensive rats increases,and electro-acupuncture at Yangming meridian points can increase the concentration of Ghrelin and NO in rat plasma,thereby activating PI3K/AKT/e NOS signal transduction pathway,protecting vascular endothelium and exerting anti-hypertensive effect.This study from the perspective of Ghrelin-regulated PI3K/AKT/e NOS signaling pathway-mediated endothelial protection,provides a new mechanism for the treatment of hypertension from Yangming,and also provides new ideas for clinical development.. |
