Therapeutic Effect Of Modified Sanhuang Guben Decoction On SLE Of Kidney-Yin Deficiency Type And Preliminary Study On Prevention And Treatment Of Glucocorticoid-Induced Osteoporosis In SLE Mice

Objective:1.To evaluate the clinical efficacy of modified Sanhuang Guben decoction in the treatment of SLE with deficiency of kidney yin;2.To explore the effect of modified Sanhuang Guben decoction on bone mineral density(BMD)of SLE patients with deficiency of kidney yin.3.Based on the theory of "kidney main bone and raw pulp",the effect of modified Sanhuang Guben decoction on bone mineral density in SLE model mice was observed,and the effect of modified Sanhuang Guben decoction on glucocorticoid-induced osteoporosis was studied.Methods:1.Clinical observation part: 32 patients with systemic lupus erythematosus(SLE)were treated with modified Sanhuang Guben decoction for a total of three months,using the method of self-control before and after taking oral corticosteroids without any change in the basis of the original oral corticosteroids for the treatment of patients with systemic lupus erythematosus(SLE).The clinical curative effects were compared before,1 month,2 months and 3 months after treatment with traditional Chinese medicine,and the femoral neck bone mineral density(BMD)was measured before and 3 months after oral administration of traditional Chinese medicine.Finally,the data were analyzed by SPSS 22.0 software.2.Animal experiment part: 30 MRL/lpr SLE model mice were randomly divided into model group,control group(prednisone group)and treatment group(prednisone Chinese medicine group)according to their weight.A total of 20 SLE model mice in the treatment group and in the control group were treated with prednisone(10mg/kg)diluted with normal saline and then fed with 0.4ml/L once a day for seven consecutive days.Seven days later,the SLE of mice was relieved and entered chronic remission stage.At this time,the treatment group was treated with prednisone(10mg/kg)and modified Sanhuang Guben decoction(3g/kg),diluted with normal saline and then diluted by 0.4.The control group was given prednisone(10mg/kg)and then diluted with normal saline,then treated with 0.4ml/L,once a day,the control group was given prednisone(10mg/kg)once a day.Weight was weighed once a week after administration,and the dose was adjusted according to body weight.The 10 rats in the model group were fed in the same environment for 8 weeks.Eight weeks after the experiment,all the mice in the three groups were taken and killed by cervical vertebradislocations.Then the left femur was carefully separated and the soft tissue was removed completely.After wrapped in 37 ? normal saline gauze,the mice were placed in the EP tube and stored in the refrigerator at-80 ?.Remove and thaw before detection.Dual energy X-ray bone for small animals Bone mineral density(BMD)of mouse femur in vitro was measured by(HOLOGIC).The data were analyzed by statistical software SPSS 22.0.Results:1.Clinical observation: 1)before the treatment of SLE patients with deficiency of kidney yin and 1 month and 2 months after treatment with modified Sanhuang Guben decoction,After 3 months,there was a significant difference in bone mineral density between the two groups(P < 0.05).2)there was a significant difference in bone mineral density before and after treatment with Chinese medicine(P < 0.05).3 months later,there was a significant difference in bone mineral density between the two groups(P < 0.05).2.Animal experiment part: model group,control group,experimental group three groups of mice bone mineral density through statistical analysis found that there was a statistical difference between the two groups(P < 0.001).Conclusion:1.Jiawei Sanhuang Guben decoction can improve the symptoms of SLE patients with deficiency of kidney yin and reduce the SLEDAI score of the patients.within three months of treatment,the longer the treatment time is,the more significant the curative effect is.2.After using Jiawei Sanhuang Guben decoction in the treatment of SLE patients with deficiency of kidney yin with glucocorticoid,the bone mineral density of the neck of the femurs was improved compared with that before the use of traditional Chinese medicine.Therefore,to a certain extent,this prescription may be able to improve the bone mineral density of patients with SLE to prevent the occurrence of osteoporosis after taking glucocorticoids.3.Jiawei Sanhuang Guben decoction improved the bone mineral density of SLE mice after glucocorticoid use,and verified that Jiawei Sanhuang Guben decoction could prevent and treat steroid-induced osteoporosis in SLE mice under the guidance of the classical theory of "kidney main bone,pulp generation".
