Objective:By consulting the ancient and modern medical literature,combined with traditional Chinese medicine theory and modern pharmacological research to determine the acaricidal formulae,entrusted Jiashan biomedicine to produce two kinds of acaricidal eye patch,which are low and high concentration respectively,low concentration acaricidal eye patch group and high concentration acaricidal eye patch group are compared with the blank group of guinea pigs in conjunctival hyperemia,corneal fluorescein staining,conjunctival impression cytology examination,and skin irritation reaction with the blank group,to assess the toxicity of topical application of acaricidal eye patch,and to evaluate the safety of topical application of low concentration acaricidal eye patch and high concentration acaricidal eye patch;a social behavior questionnaire survey of patients with ocular demodex infection to assess infection-related factors.Methods: 1.Animal experiment Thirty-two healthy guinea pigs were randomly divided into blank group,low concentration acaricidal eye patch group,high concentration acaricidal eye patch group,and skin irritation experiment group,with 8 rats in each group.There was no treatment in the blank group;low concentration acaricidal eye patch group,high concentration acaricidal eye patch group daily time eye patch applied to each eye,each time 30 min,once a day,the experimental period was 14 days;in the skin irritation test group,the both sides of the guinea pig 'hair was polled.The left side was coated with saline gauze and covered with oiled paper to moisturize,and the right side was applied with high concentration eye patch.After 4 hours,the reagents on both sides were removed,and the skin reactions of the test sites were observed after 1,24,48,and 72 hours.2.Questionnaire Thirty-seven patients with ocular Demodex infection in the outpatient department of the Eye Hospital of Chengdu University of Traditional Chinese Medicine were collected as subjects in the study.Their questionnaires were used to record their gender,age,place of residence,facial cleansing method,number of cleansing times per day,skin type,pets or not is statistical analysis,etc.Results: 1.Animal experiment(1)Conjunctival hyperemia degree:There was no statistical difference in the blank group and low concentration acaricidal eye patch group before and after the experiment(P>0.05),and the difference of high concentration acaricidal eye patch group was statistically significant(P<0.05),there was significant difference in the high concentration group before and after the experiment(P<0.05),there were no significant differences between the groups before and after the experiments(P>0.05);(2)Corneal fluorescein staining: There was no statistical difference in the blank group,low concentration acaricidal eye patch group and high concentration acaricidal eye patch group(P<0.05);The difference between the three groups was statistically significant after the experiment(P<0.05);(3)Conjunctival impression cytology: There was no statistically significant difference between the three groups before and after the experiment.There was no significant difference before and after the experiment in the three groups(P>0.05).(4)The skin irritation test group did not find skin erythema and edema after 1,2,48,and 72 hours after removing the bilateral reagents on the back of the guinea pig.2.Questionnaire The proportion of female patients with ocular demodex infection was 62.16%,the proportion of males was 37.84%;the youngest was 18 years old,the oldest was 81 years old,the average age was 41.51±18.52 years old;those living in cities accounted for 78.38%,those living in rural areas accounted for 21.62%;those who used warm water for cleansing accounted for 59.46%,those who used cold water accounted for 40.54%;those who used skin care products regularly accounted for 43.24%,those who seldom used skin care products accounted for 56.76%;those who used household cleaning products strictly for single use accounted 32.43%,sharing accounted for 67.57%;skin type was dry accounted for 10.81%,oily accounted for 32.43%,mixed type accounted for 56.76 %;pets accounted for 16.22%,and those who denied pets accounted for 88.78%.Conclusion: 1.High concentration acaricidal eye patch led to conjunctival hyperemia;2.Low concentration acaricidal eye patch without adverse reactions such as conjunctival hyperemia,corneal epithelial loss,conjunctival cytology changes,skin irritation,etc;3.Skin type,and shared brushing supplies are related factors of ocular demodex infection.
