Objective: Acne vulgaris is one type of chronic inflammatory skin disease.TIMP-2 is an important enzyme in the degradation of extracellular matrix,it can specifically inhibit the activity of matrix metalloproteinase-2(MMP-2).MMP-2 and TIMP-2 play important role in the balance of extracellular matrix synthesis and degradation metabolism.Enhanced MMP-2 activity or decreased TIMP-2 activity will destroy the dynamic balance of ECM.And it will promote the occurrence and development of inflammation.Acne,as an inflammatory disease,is closely related to proinflammatory factors such as MMP-2 and TIMP-2.In recent years,the relationship between MMP-2 and TIMP-2 polymorphisms and the incidence of acne has attracted more and more attention.Studies have shown that the single nucleotide polymorphisms(SNP)at MMP-2 rs243865 and TIMP-2 rs8179090 may be associated with the risk of acne in some populations.In this research,the SNP of MMP-2 and TIMP-2 in patients with acne vulgaris in Han Chinese of Hebei province were studied to verify the susceptibility genes of acne vulgaris in different ethnic groups.Methods:1.Patient group included 106 individuals(45 males and 61 females)with acne vulgaris.The control group included 120 healthy individuals(52 males and 68 females).2.After extracted the peripheral venous blood 4mL from every individual,Glucose(GLU),Triglyceride(TG)and Testosterone(T)were detected and the whole blood genomic DNA were extracted.The target gene was obtained.Then we detected the SNP of the MMP-2 gene at position rs243865 and TIMP-2 gene at position rs8179090 of the acne vulgaris patients and healthy controls by the method of genetic sequencing.Results:1.The average level of fasting glucose(GLU)was respectively 4.86±0.55mmol/L and 4.91±0.61mmol/L in the patient and the control group.There was no significant difference between the two groups.2.The average level of triglyceride(TG)was respectively 1.09±0.65mmol/L and 1.23±0.84mmol/L in the patient and the control group.There was no significant difference between the two groups.3.The average level of testosterone(T)was respectively 17.47±2.62nmol/L and 2.94±0.52nmol/L in the male patient and the male control group.There was no significant difference between male patient and male control group.The average level of testosterone(T)was respectively 2.94±0.52 nmol/L and 1.11±0.31nmol/L in the female patient and the female control group.There was significant difference between female patient and female control group.4.The detection of MMP-2 rs243865 gene polymorphism showed three genotypes: CC、CT and TT.In the patient group,the CC genotype was 59 cases(55.66%),the CT genotype was 42 cases(39.62%),the TT genotype was 5 cases(4.72%).In the control group,the CC genotype was 63 cases(52.5%),the CT genotype was 49 cases(40.83%),the TT genotype was 8 cases(6.67%).The CT+TT genotypes were 47 cases(44.33%)in the patient group and 57 cases(47.5%)in the control group.There was no significant difference between the two groups.5.The detection of TIMP-2 rs8179090 gene polymorphism showed three genotypes: GG、GC and CC.In the patient group,the GG genotype was 68 cases(64.15%),the GC genotype was 32 cases(30.19%),the CC genotype was 6 cases(5.66%).In the control group,the GG genotype was 110 cases(91.67%),the GC genotype was 10 cases(8.33%),the CC genotype was 0case(0%).The GC+CC genotypes were 38 cases(35.85%)in the patient group and 10 cases(8.33%)in the control group.The GC+CC genotypes were statistically difference between the two groups.Conclusion:1.GLU and TG were not associated with acne vulgaris.2.The increase of T level may be related to the female patients of acne vulgaris.3.The SNP at the rs243865 position of MMP-2 gene was not associated with acne vulgaris.4.The SNP at the rs8179090 position of TIMP-2 gene may be associated with acne vulgaris,and the presence of the C allele at the rs8179090 position of TIMP-2 gene may increase the risk of acne vulgaris.
