Objective To study the extraction technology of essential oil of A.nobilis in Heilongjiang and analysis the main components of its essential oil and floral water,and to compare it with A.nobilis in Xinjiang and Matricaria recutita to analyze the quality difference.The antimicrobial activity of essential oil and floral water of A.nobilis in Heilongjiang was determined by in vitro antimicrobial experiment,to compared that with A.nobilis in Xinjiang and Matricaria recutita.It provides a reference for the further development of A.nobilis in Heilongjiang Province.Methods The optimum extraction method of essential oil of A.nobilis in Heilongjiang was determined by single factor experiment and orthogonal experiment.The essential oil of A.nobilis was refined by anhydrous sodium sulfate drying method,vacuum distillation method and ether extraction method,the best method for the refinement of essential oil of A.nobilis was screened by component comparison after GC-MS.The main components of essential oil and floral water of A.nobilis in Heilongjiang was tested by GC-MS,and compare with A.nobilis in Xinjiang and Matricaria recutita.The antimicrobial activities of essential oil and floral water of A.nobilis in Heilongjiang were analyzed by method of filter paper,and compared with A.nobilis in Xinjiang and Matricaria recutita to analysis the difference of antimicrobial activity.Result The results showed that the optimum extraction conditions of essential oil of A.nobilis in Heilongjiang were as follows: ratio of material to liquid 1:12,soaking time 12 h and extraction time 3 h.The main components of essential oil refined by anhydrous sodium sulfate drying,vacuum distillation and ether extraction method had little difference.The esters of essential oil refined by anhydrous sodium sulfate drying were 64.0%,the esters of essential oil refined by vacuum distillation were 67.0%,and the esters of essential oil refined by ether extraction were 68.0%,and the main components are alpha-pinene,3-methyl-2-butenoic acid-3-methyl butyl ester and cyclopropane carboxylic acid-3-methyl butyl ester.The esters accounted for 66.67% in the essential oil of A.nobilis in Heilongjiang,and the main components were alpha-pinene,3-methyl-2-butenoic acid-3-methyl-butyl-2-ene ester and pinocarvone.The esters accounted for 69.23% in the essential oil of A.nobilis in Xinjiang,the main components were the same as those of A.nobilis in Heilongjiang,but the contents were quite different.Olefins accounted for 50.00% of the essential oil of Matricaria chamomilla,while esters accounted for only 9.09%,the main components were alpha-bisabolol oxide B and chamazulene,etc.The esters accounted for 68.75% in floral water of A.nobilis in Heilongjiang,and the main components were 3-methyl-2-butenoic acid-3-methyl butyl ester and pinocarvone.The esters accounted for 70.00% in floral water of A.nobilis in Xinjiang,and the main components were 3-methyl-2-butenoic acid-2-pentyl ester and butyric acid-3-methyl-butyl-2-ene ester,which were quite different from those in floral water of A.nobilis in Heilongjiang.The esters accounted for 63.64% in floral water of Matricaria chamomilla,and the main components were alpha-pinene and 3-methyl-2-butenoic acid-3-methyl butyl ester.The order of MIC of essential oil and floral water of A.nobilis in Heilongjiang was Staphylococcus albicans > Escherichia coli ? Bacillus subtilis = Staphylococcus aureus.The antimicrobial effect of essential oil and floral water of A.nobilis in Xinjiang was slightly higher than that of A.nobilis in Heilongjiang.This may be due to the high content of pinocarvone,2-methyl butyl isobutyrate and 3-methyl-2-butenoic acid-2-pentyl ester in essential oil and floral water of A.nobilis in Xinjiang.The antimicrobial effect of essential oil and floral water of Matricaria recutita had slightly higher than essential oil and floral water of A.nobilis,this may be due to the higher content of alpha-Bisabolol oxide B,chamazulene and alpha-pinene in essential oil and floral water of Matricaria chamomilla,while A.nobilis essential oil and floral water had no or less.The study found in essential oil and floral water of A.nobilis in Heilongjiang compatibility with essential oils and floral water of Chrysanthemum and Oimum gratissimum,when the ratio of essential oil and floral water of A.nobilis to Chrysanthemum and O.gratissimum L.was 1:2,the antimicrobial activity was the best.Conclusions The main components of essential oil and floral water of A.nobilis in Heilongjiang are esters,which have high antimicrobial activity to Staphylococcus albicans and Staphylococcus aureus.Due to regional differences,the main components and antimicrobial activity of essential oil and floral water of A.nobilis in Heilongjiang are quite different from those of A.nobilis in Xinjiang.Because of the different species,the chemical composition and antimicrobial activity of essential oil and floral water of A.nobilis and Matricaria recutita are quite different.
