Objective To synthesize and characterize Gallic acid Oxyvanadium Complexes?GAOV?,optimize the synthesis process,investigate the acute toxicity of GAOV,explore its antioxidant activity in vitro,hypoglycemic activity in vitro and in vivo.Methods GAOV was prepared by using gallic acid as ligand and vanadyl sulfate as the source of vanadium.The effects on the yield of reaction time,pH,temperature and feed ratios were investigated.The orthogonal experimental design method was used to optimize the synthesis process of GAOV.The GAOV was characterized by UV-vis,DSC,IR,XRD and TGA.Disposable oral toxicity was tested of GAOV and vanadyl sulfate according to the national acute toxicity test standard.UV-spectrophotometry was used to determine the scavenging ABTS,H2O2 and DPPH free radicals of GAOV.The scavenging rate was determined of Hydroxyl by salicylic acid method.The inhibition effect on?-amylase and?-glucosidase were mensurated of GAOV by DNS colorimetry and PNPG substrate method.Diabetic mouse model was established by streptozotocin?STZ?.The animals were divided into control,model,positive group,and high,medium and low dose groups of GAOV and vanadyl sulfate,and the changes of diet,water intake,body weight and the FBG of mice were monitored.The UREA,Cr,ALB,ALT,CHO,TG,LDL-C,HDL-C of each group were measured.The indexes of pancreas,liver,kidney and spleen were calculated and liver,kidney and pancreas tissues were observed by HE.Results The optimum synthesis process of GAOV were 70?,pH=7,10 h,the molar ratio was 1:1.The results of acute toxicity test showed that the LD50 of GAOV>3000 mg·kg-1,which was much smaller than that of vanadyl sulfate.The IC50 value of Vc,Gallic acid and GAOV for scavenging ABTS were 0.009 mg·mL-1,0.017 mg·mL-1,0.048 mg·mL-1;for scavenging H2O2were 0.0160 mg·mL-1,1.583 mg·mL-1,0.128 mg·mL-1;for scavenging·OH were 0.0176mg·mL-1,0.0285 mg·mL-1,0.958 mg·mL-1;for scavenging DPPH were 0.0002 mg·mL-1,0.00386 mg·mL-1,0.0168 mg·mL-1.The inhibiting results of?-amylase were acarbose>GAOV>gallic acid,and the inhibition of?-glucosidase were GAOV>acarbose>gallic acid.The results of hypoglycemic test in vivo showed that there were significant differences in body weight,dietary water consumption,FBG,serum biochemical indicators,and blood lipids in the GAOV group?P<0.05?,compared with the model group,and there was no significant difference between the GAOV medium dose group and positive group?P>0.05?.Conclusions The preparation method of GAOV was simple,safe,reliable,with stable yield and good repeatability.Compared with vanadium sulphate,the toxicity of GAOV was greatly reduced,and the GAOV had a strong scavenging effect on free radicals such as ABTS,H2O2 and DPPH in vitro.It had a strong inhibitory effect on?-amylase and?-glucosidase.The GAOV has a certain regulatory effect on blood glucose and serum indexes in diabetic mice induced by STZ.This study provided new ideas for further research on vanadium oxygen complexes.
