Objective:The body reacts rapidly and strongly after being exposed to acute radiation,and cells,tissues and organs all cause varying degrees of damage,and even direct death in severe cases.The experimental study showed that compound raspberry seed powder had certain protective effects on peripheral blood leukocytes,platelets,lymphocyte DNA and liver of rats with acute radiation injury.Bone marrow cells are the general name of all kinds of cells in bone marrow.As the cornerstone of human hematopoiesis,immunity and wound repair,their research is of great significance.Compound raspberry seed powder on acute radiation injury mice bone marrow cells,this experiment from cellular level to acute radiation injury mice number of nucleated cells of bone marrow cells,chromosome mutation,apoptosis,and so on and so forth were studied,discussed the compound raspberry seed powder aqueous solution on the effect of acute radiation injury mice bone marrow cells and protection.Methods:190 mice were divided into blank control group,model control group and compound raspberry seed powder solution treatment group(low/medium/high dose group).In addition to the blank control group,mice in the other groups were given the subjects(distilled water or compound raspberry seed powder solution with different concentrations)for 15 days after gavage,followed by one-time whole-body X-ray irradiation,and on the third day after the X-ray irradiation,all mice were killed as required and the obtained bone marrow cells were tested experimentally.The total number of nucleated cells in the suspension of bone marrow cells of mice in each group was compared by counting under the microscope.The chromosome variation of bone marrow cells in each group was determined by Giemsa staining.The apoptosis of bone marrow cells in each group was detected by PI single staining assay.Rhodamine123 staining was used to detect the variation of mitochondrial membrane potential of bone marrow cells in each group.Results :(1)In the detection of the number of nucleated cells in bone marrow cells,the model control group decreased by 58.5% compared with the blank control group after irradiation(P<0).01).Compared with the model control group,the medium and high dose groups of compound raspberry seed powder increased by 78.2% and 116.9%,respectively(P <0.01).(2)In the detection of micronucleus rate of bone marrow cells,compared with the blank control group,the micronucleus rate of model control group increased by 230.0% after irradiation(P <0.01).Compared with the model control group,the micronucleus rate of bone marrow cells in mice exposed to high dose compound raspberry seed powder decreased by 27.3%(P <0).01).In the detection of bone marrow cell cycle,compared with the blank control group,the changes in other groups were significant(P <0.01).Compared with the model control group,the proportion of cells in G1 phase and S phase in the medium dose group increased in the high dose group(P < 0.05),and the proportion of cells in G1 phase increased significantly in the high dose group(P < 0.01).(4)in the bone marrow cell mitochondrial membrane potential detection,compared with the blank control group,the other groups changed significantly(P <0.01).Compared with the model control group,mitochondrial membrane potential of bone marrow cells was significantly increased in the medium and high dose groups(P < 0.01).Conclusion:The results showed that the bone marrow cells of the mice were obviously damaged after receiving a certain dose of X-ray.Compound raspberry seed powder can protect the body from acute radiation injury in a dose-dependent manner.Compound raspberry seed powder can increase the number of bone marrow cells,reduce the micronucleus rate of bone marrow cells,increase the mitochondrial membrane potential of bone marrow cells,and reduce the cell cycle arrest of bone marrow cells,so as to inhibit the apoptosis of bone marrow cells in irradiated mice,which may play an important role in the protection against bone marrow cell damage caused by radiation.
