Intestinal microbiota are closely related to digestion and absorption of the body,development and function of the immune system.However,intestinal microbiota are susceptible to host genes and the external environment.In particular,the use of antibiotics seriously threatens the homeostasis of intestinal microbes.Long-term abuse of antibiotics can change the number and composition of intestinal microbes and result in intestinal microbiota dysbiosis.Therefore,it is important to explore how to eliminate intestinal microbial disorders caused by antibiotics and other side effects.Ginsenoside,as the main active ingredient of ginseng herb,can exert good pharmacological,and has a certain role in regulating intestinal microbiota.Protopanaxatriol-type saponin(PPT),as one of the main components of ginsenosides,has good anti-tumor and anti-diabetes effects,so it is of great significance to study its regulatory effect on intestinal bacteria.Therefore,the purpose of this study is to understand the regulatory effect of PPT on intestinal bacteria of C57/BL6 mice.In this study,we constucted model mice which have disorder intestinal bacteria by giving 300 mg/kg body weight of cephalosporin through intragastric administration.And then,gave the model mice 50,100,25 mg/kg body weight PPT to see the effects of PPT on mice disordered intestinal bacteria,colonic inflammation and the damage of intestinal barrier integrity caused by antibiotics.The 16 S r RNA gene comprising V3-V4 regions were analyze the abundance and composition of micobiota in mice feces in different treatment groups.The concentration of short-chain fatty acids(SCFAs)in the feces of mice in different treatment groups were analyzed by GC-MS technology.The expression levels of inflammatory cytokines(TNF-?,Il-1?,Il-6,IL-1?,IFN-?)and tight junction protein(ZO-1,occludin)in colon tissues of mice in different treatment groups were analyzed by RT-q PCR.Western blot(WB)was used to analyze the expression of inflammatory cytokines,short-chain fatty acid receptor proteins(GPR109A,GPR43,GPR41)and TLR4 signaling pathway related proteins(TLR4,JNK,NF-?B)in colon tissues of mice from different treatment groups.The results showed that the use of antibiotic caused the intestinal bacteria dysbiosis and colonic inflammation,destroyed the integrity of intestinal barrier,reduced the concentration of SCFAs and the expression of short chain fatty acids receptor proteins.However,the PPT improved the gut microbiota composition,decreased the inflammation,and increased the concentration of short-chain fatty acids as well as tight junction proteins in antibiotic-treated mice.These findings indicate that PPT has a protective effect on intestinal microbes,and enhances the integrity of the intestinal barrier as well as alleviates colonic inflammation in antibiotic-treated mice.
