Objective:Observe the effect of 1,25?OH?2D3 on the expression of mTOR in HK-2 cells stimulated by TGF-?1 and the role of 1,25?OH?2D3 in epithelial-mesenchymal transition of HK-2 cells.Methods:1.Selected HK-2 cells and give TGF-?1 stimulation,optimize conditions of TGF-?1 stimulate for HK-2 cells based on the?-SMA expression using Real Time-PCR analysis.2.Groups:normal control group,TGF-?1?5ng/ml?group,TGF-?1+1,25?OH?2D310-10mol/l group,TGF-?1+1,25?OH?2D310-9mol/l group,TGF-?1+1,25?OH?2D310-8mol/l group,Co-cultivation for 48h.The protein expressions of P-mTOR,?-SMA and E-Cadherin were detected by Western blot.Results:1.According to?-SMA gene expression,HK-2 cells was treated with TGF-?1?5ng/ml?to induce EMT.2.Compared with the normal control group,the expression of P-mTOR was increased after TGF-?1 stimulation,which significantly reduced in TGF-?1+1,25?OH?2D3,but it still higher the normal control group,the difference was statistically?P<0.005?.3.Compared with the normal control group,the expression of?-SMA was increased and the expression of E-Cadherin was decreased after TGF-?1 stimulation.After treatment with 1,25?OH?2D3,the expression of?-SMA was lower than TGF-?1 group and higher than normal control group,the expression of E-Cadherin was higher than TGF-?1 Group and lower than normal control group,the difference was statistically significant?P<0.005??P<0.005?.Conclusion:1.1,25?OH?2D3 attenuates epithelial-mesenchymal transdifferentiation induced by TGF-?1 in HK-2 cells;2.1,25?OH?2D3 attenuates epithelial-mesenchymal transdifferentiation may be associated with the mTOR pathway..
