The zika viru is a teratogenic yellow virus transmitted by mosquitoes,which is linked to the birth defects of a newborn and the Barli syndrome of adults,which can also be transmitted through sex.Recent research on zika virus(ZIKV)find it can be hidden in male genitals in more than 6 months,which increases the burden of the control of ZIKV and poses a potential threat to human health.There is little research about supporting viral replication and existence in the human testes.Existing studies have focused on the extracorporeal infection of the sertoli cells,as well as the research of rodents and primate models,and the sertoli cells have important regulation and protective effects on male reproductive functions,and are an unsubstitutable type of cells in the testicles;And it is also an important target cell of ZIKV infection,so it is of guiding significance for the follow-up of ZIKV infection from the testicular support cells.However,the availability and use of the primary testicular sertoli cells is indisable,and the establishment of a rodent model is an anti-processing that may be not able to be convincible.The primate model is similar to the human disease,but the cost is high,so there is a requirement for an infection that is similar to the human,the convenient,low-cost experimental animals as the study of the ZIKV latent infection,to explore the basic laws of ZIKV and its possible mechanisms for male infections.In this study,we use the tree shrew,which was similar to the human being,was established by the two-step enzyme digestion method and the time change fluid,and the mature original cell of the epidites was supported by the external separation and cultivation of the cell body,and the separate cells were detected by the antibody didyeing,and the high quality cells were obtained to be used for external infection.To detect the changes of viral load in infected cells by qRT-PCR,and to explore the normal number of ZIKV infection levels in the cell level;Based on the real-time fluorescence quantitative PCR combined with immunofluorescence technology,an important receptor AXL,which is closely related to the infection of the yellow virus,was used to explore the correlation between the cell model and the ZIKV infection in the cell model,and the role that could be done.Finally,through qrt-pcr test,the infection pattern of ZIKV in the body experiment was detected,and the expression of the CD4+and IFN gamma was found by flow cell count and ELISA detection,and the following results were obtained:1)As an experimental animal,tree shrews are more convenient and easier to obtain and purify in the isolation and culture of testicular supporting cells than existing animals;2)High expression of ZIKV can be maintained for 12 days in SC in vitro infection,and AXL may be related to ZIKV infection as an important receptor,but AXL is not the main receptor for ZIKV entry in the testicular supporting cells of tree shrew as in the existing rodent model.3)In healthy 5-month old male tree shrews infected with ZIKV,viremia occurred 3 days after the infection,causing a small number of organs such as the spleen and liver histopathological changes,and was highly expressed in the testis and epididymis on the second day.Later,no virus expression was detected in the body,and no latent phenomenon occurred.4)The number of CD4+T cells increased in infected tree shrews on the 3rd and 5th days,and the expression of IFN-increased significantly on the 5th day,which may be involved in the process of virus clearance in the body through adaptive immune pathways.This study,using protocell separation and culture to develop an external infection,observed that the testicles supported cells were highly sensitive to the zika infection,which could be associated with the tam family receptor AXL.In the internal infection,the replication of the male tree will not appear as the latent phenomenon of the male tree shrew,and the abnormal high expression of CD4+T and IFN-y.IFN-y is a key point in the failure of latent phenomena in the middle of the infection.These findings provide important insights into how the zika virus can survive in male reproductive tract,which in effect may help develop antiviral treatments or strategies to minimize the spread of the pathogen.
