Objective 1 To explore the role of the mediating health care theory in the pathogenesis of airway hyperresponsiveness through the airway hyperresponsiveness rat model induced by PM2.5.2 Guizhi Pingfeng decoction was used to intervene in the airway hyperresponsiveness rat model induced by PM2.5,to study the efficacy of this prescription and the therapeutic mechanism based on Th17/Treg immune balance.MethodA total of 48 Wistar rats were randomly divided into a blank control group,a PM2.5 model control group,a Yupingfeng group,a low dose Guizhi Pingfengtang group,a medium dose Guizhi Pingfengtang group and a high dose Guizhi Pingfeng group.PM2.5 animal models of rats were constructed by tracheal aspiration.Yupingfeng decoction and three doses of Guizhi Pingfeng decoction were applied to each group for gastric intragastric therapy.The blank control group and the PM2.5 model control group were given the same amount of normal saline for 28 consecutive days.Through the detection of blood IL-17,IL-6,IL-10,TGF-?and other cytokines,the pathological changes of airway inflammation and lung tissue morphology and structure were observed under light microscope,and the improvement of Guizhi Pingfeng decoction on PM2.5 induced inflammation was observed.By detecting the mRNA and protein expressions of IL-17 and Foxp3 in lung tissues,the regulatory effect of Guizhi Pingfeng decoction on Th17/Treg imbalance was investigated.ResultCompared with the blank group,model group of the lung tissue of rats induced by PM2.5 apparent inflammatory change,alveolar disorder,bronchial secretions increase,cilia clutter and fall off,hyperplasia of goblet cells,the protein expression in the rat Foxp3 mRNA reduce,IL-17 higher protein expression of mrna,BALF of neutrophils,macrophages,and lymphocytes increased significantly,the number of serum IL-6,IL-17 levels drop,TGF-?,the higher content of IL-10,occurrence of Th17/Treg imbalances.After the drug intervention,compared with the model group,rats in each dose of Guizhi Pingfeng decoction showed different degrees of inflammatory improvement in lung tissue,alveolar recovery,cupping cell reduction,decreased expression of inflammatory factors,decreased number of inflammatory cells in BALF,decreased levels of IL-6 and IL-17 in serum,and increased levels of TGF-? and IL-10.Conclusion 1 Guizhi Pingfeng decoction can effectively regulate Th17/Treg imbalance by down-regulating IL-6 and IL-17 and up-regulating IL-10 and TGF-?,so as to reduce the secretion of inflammatory cells and the release of inflammatory mediators,reduce airway inflammation,and achieve the purpose of treating airway hyperresponsiveness.2 Guizhi Pingfeng decoction can reconcile Yingwei,nourish the lung Yin,enrich the Qi and firm the surface,and effectively improve the symptoms caused by PM2.5,so as to achieve the purpose of treating airway hyperresponsiveness..
