Objective:Sorting out the historical evolution and development of Polygoni multiflori and its different processed products,combined with the research status of Polygoni multiflori artillery products,experimental research on the toxicity and hepatotoxicity of Polygoni multiflori and its different processed products,to evaluate the toxic effects of different processed products of Polygoni multiflori based on the whole and in vitro levels.Experimental study to investigate the toxicity characteristics,damage degree and recovery of Polygoni multiflori and its processed products,and evaluate the toxicity of isolated liver,and provide certain experimental basis for the toxicity and hepatotoxicity evaluation of Polygoni multiflori and its different processed products.Methods:According to the local processing specifications,different pieces of Polygoni multiflori were prepared on the basis of Sample 1?Raw?,followed by Sample 2?steamed 6h?,Sample 3?Bean juice-6 h?,Sample 4?Bean-processing for 3 h?,Sample 5?Bean juice and yellow wine-18 h?,Sample 6?black bean juice separated by water stew 6 h?,Sample 7?black bean juice Polygoni multiflori cooked for 12h?,Sample 8?black bean Polygoni multiflori cooked for 12h?.According to the classical acute toxicity test method,the acute toxicity study of the drug samples Sample 1,Sample 2,Sample 3,Sample 4,Sample 5,and Sample 6 was performed;and the drug samples Sample 1,Sample 2,Sample 3,Sample 4Sample 5,Sample 6 were taken on the liver toxicity"quantity-time-toxic"relationship in mice,observe the death and toxicity of mice after administration,check the serum levels of ALT,AST,TBA,TBIL,AKP after drug administration,The"quantity-toxin"experiment adds IL-2 and TNF-?indicators,calculates the organ coefficients of liver,spleen,lung,kidney,adrenal gland and thymus,takes the liver for histopathological examination,and uses long-term toxicity evaluation method to carry out the medicine.Samples Sample 1,Sample2,and Sample 3 were tested for long-term toxicity in normal rats.The general conditions such as body weight,daily diet,and daily drinking water were observed after administration and recovery.Blood routine,blood biochemical indicators,and dirty Detection of indicators such as organ coefficient and histopathology;using liver perfusion technique to study the Sample 6 were calculated according to the raw drug amount of 345.60 g·kg-1·d-1,328.32 g·kg-1·d-1,309.60 g·kg-1·d-1,354.24 g·kg-1·d-1,336.96 g·kg-1·d-1;"Time-toxic"results showed that serum ALT,AST,TBA,TBIL,AKP levels peaked 2 h after drug administration.The"quantity-toxicity"relationship study showed that compared with the normal group,the water extract of Polygoni multiflori can cause different livers in mice.The degree of damage,serum ALT,AST,TBA,TBIL,AKP,IL-2 and TNF-?activity showed different degrees of significant difference;continuous 90-day long-term toxicity test results,compared with Sample 1,Sample 2 and Sample 3 The levels of ALT,BUN and TG in the blood of the rats were significantly decreased,and the ALB levels in Sample 3 were significantly increased.There was no significant difference in the blood biochemical parameters between Sample 2 and Sample 3.The pathological results showed that the long-term administration of Sample 1 could cause large In the male reproductive organs,Sample1 and Sample 2 showed lung injury,and the ALT and GGT levels of Sample 3 decreased significantly after 45 days of withdrawal.The results showed no male reproductive organs and lung damage;compared to the isolated liver perfusion experiment,other drug samples of various doses Sample(Sample 1:0.544.32 g·kg-1,Sample 28:1.088.64 g·kg-1)Drug sample Sample 5 high,medium dose,and Sample 6 high dose liver homogenate ALT was not significant,toxicity was significantly less than ALT significantly increased drug samples Sample 1,Sample 2,Sample 4,Sample 7 and Sample 8.Conclusion:The acute toxicity of Polygoni multiflori and its different processed products to mice is:Sample 1>Sample 4>Sample 3>Sample 6>Sample 2>Sample 5,Polygoni multiflori can cause significant toxicity to mice;single-time administration to mice A certain dose of Polygoni multiflori and its water extracts from different processed products can cause different degrees of liver damage and exhibit certain relationship between time and toxicity.The results of long-term toxicity test showed that the aqueous extracts of Sample 1,Sample 2 and Sample 3 in the 90-day continuous drug administration had certain toxicity,and the toxicity showed that the blood biochemical indexes such as ALT,AKP,GGT,ALB,BUN and CREA increased significantly,and the method of processing steamed after the black bean juice has better attenuation effect;At the level of body liver,water extracts of Polygoni multiflori and its different processed products can cause liver tissue damage,and Sample 5 has minimal hepatic toxicity.It is recommended to use black bean juice and yellow wine as auxiliary materials and mix well with Polygoni multiflori for 4 h,put it into the steamer,add some water,seal,steam for 18 h,pour the can in the middle once,until the juice is exhausted,both inside and outside are brown to dark brown,take out,dry processing method.
