Objective:This study was to investigate the correlation between the number of endothelial progenitor cells(EPCs)in peripheral blood and abdominal aortic aneurysm(AAA),thus providing a theoretical proof for the application of EPCs transplantation to AAA in later experiments.Methods:1.Groups:Experimental group:patients with AAA in the First Affiliated Hospital of Soochow University from June 2018 to October 2018,(n=7).All patients were confirmed as AAA by vascular CTA based on over 50%of abdominal aorta diameter than the normal group.Control group:normal bdominal aorta diameter according to the principle of matched group(n=7).2.Clinical information:The date included matching age,sex,blood pressure and blood sugar.3.The number of EPCs:Mononuclear cells were collected by density gradient centrifugation and stained via CD34-FITC and CD309-PE,then detected the number of positive cells by flow cytometry.4.AAA mice and rat models were induced to validate the correlation between the number of endothelial progenitor cells in peripheral blood and abdominal aortic aneurysm.Results:1.Clinical information:Experimental group included 5 males and 2 females.the average age of male patients was 65 years old,the average age of female patients was 74 years old.Control group included 4 males and 3 females.The average age of male is 68 years,and the average age of female is 73 years.2.The number of EPCs:EPCs in peripheral blood from patients with AAA(0.874±0.129)was slightly decreased than that in concrol group(1.420±0.289)(P<0.01).Two weeks after AAA endovascular aortic repair,the number of EPCs(1.573±0.574)in peripheral blood more than that control group(0.874±0.129)(P<0.05).3.Rat AAA models induced by elastase displayed fewer number of EPCs in peripheral biood(0.875±0.377 VS 1.873±0.840 p<0.01).Mouse AAA model via angiotensin-combinated with a high-fat diet displayed no significant statistical difference(1.532±0.498 VS 1.860±0.833 p>0.05).Conclusion:1.Clinical data indicate that the number of EPCs in peripheral blood fom AAA patients was reduced.EPCs was rescued after clinical endovascular repair.2.The number of EPCs was consistently decresesd in rat AAA by elastase treatment.3.The number of EPCs was no significant changes in mouse AAA induced by angiotensin-combinated with high-fat diet.
