Stat5(Signal transducer and activator of transcription 5)is an important member of the signal transduction and activator of transcription(Stat)family.Stat5 has two subtypes,Stat5 a and Stat5 b,both of which are highly derived.And they are similar in function,mainly involved in regulating the growth and development of the breast,cell proliferation and apoptosis.In addition,it plays an important role in the process of tumor development,proliferation and metastasis.Delta5 Stat5 a is a variant of Stat5a(in order to distinguish it from Delta5 Stat5 a,the original Stat5 a is called FL Stat5a),which was first cloned and reported by Tan et al.,which is the product of alternative splicing of mRNA precursor of Stat5 a gene expression product.The 90 nucleotide sequence corresponding to exon 5 in the mature mRNA is deleted,resulting in the deletion of the N-terminal 30 amino acid residues of the corresponding protein product.To study the effect of overexpressing Delta5 Stat5 a on the transcriptome of breast cancer cells;Methods: we can constructe the FL Stat5 a and Delta5 Stat5 a lentiviral stable cell lines,and then,extract the total RNA from stable cells,make it isolate and purify.The sequencing data was then obtained by high-throughput sequencing technology-RNA sequencing(RNA-seq)and analyzed using bioinformatics and statistics.RESULTS: 1)The number of genes in FL Stat5 a were 22,319,the number of genes in?5 Stat5 a were 21,904,and the number of genes in con were 23,960.2)overall level analysis of transcriptional expression:Delta5 Stat5a(23044),FL Stat5a(23330),con(25688);3)the number of mRNAs in con were 16334,The number of mRNAs in Delta5 Stat5 a were 15,184,and the number of mRNAs in FL Stat5 were 15,364.Delta5Stat5 a compared with FL Stat5 a,significantly different mRNA has 23,up-regulated mRNA inDelta5 Stat5 a has 17,and down-regulated mRNA has 6.4)The number of ncRNAs in con were 9354,the number of ncRNAs in?5 Stat5 a were 7860,and the number of ncRNAs in FL Stat5 a were 7966.Delta5 Stat5 a compared with FL Stat5 a,significantly different ncRNA has 12,up-regulated mRNA in Delta5 Stat5 a has1,and down-regulated mRNA has 11.5)The number of new transcripts ncRNA in con were 104,the number of new transcript ncRNA in?5 Stat5 a were 232,and the number of new transcript ncRNA in FL Stat5 a were 346.6)KEGG pathway enrichment analys showed that the target gene of differentially expressed ncRNA of FL Stat5 a and Delta5Stat5 a,were mainly enriched in cancer-related pathways: p53 signaling pathway,NF-kappa B signaling pathway.7)GO function analysis showed that the target gene of differentially expressed ncRNA of FL Stat5 a and Delta5 Stat5 a,was mainly enriched in the biological process:The ribonucleoprotein complex biological origin,nuclear transcription mRNA catabolism,cell composition tissue or biogenesis,mRNA acid decomposition Process,ribonuclease decomposition process,processing and metabolicprocesses of rRNA,ribosome biosynthesis,major metabolic processes,cellular metabolic processes;Enriched in Cell component:organelles,intracellular ribonucleoprotein complex,intracellular part,cytoplasmic ribosome,part of intracellular organelle;Enriched in Molecular function:RNA binding,histone acetyltransferase activity(H4-K5 specificity,H4-K8 specificity,H4-K16 specificity),H4 histone acetyltransferase activity,ribosome structural composition,transferase activity,acyl group other than amino group transfer,structural molecular activity,transferase activity,transfer acyl group,etc.Conclusion: 1)Overexpression of FL Stat5 a and Delta5 Stat5 a genes can affect gene expression in MCF-7 breast cancer cells.2)Overexpression of FL Stat5 a and Delta5 Stat5 a genes,the expression profile and coverage of stably transfected cells changed,and the main changes in expression levels were chromosomes 1,11,and 16.3)Delta5 Stat5 a gene can promote the proliferation of breast cancer epithelial cells.4)KEGG pathway enrichment analysis showed that the target gene of differentially expressed ncRNA of FL Stat5 a and Delta5 Stat5 a,were mainly enriched in cancer-related pathways: p53 signaling pathway,NF-kappa B signaling pathway.Through p53 signaling pathway and NF-kappa B signaling pathway,maybe cell proliferation and metastasis are affected.This study provides important data support for further exploring the molecular mechanisms of FL Stat5 a and Delta5 Stat5 a in human breast cancer cells,and lays a solid foundation for a deeper understanding of the role of FL Stat5a and Delta5 Stat5a.
