Purpose:To explore the effect of XPLN on the biological activity of human colon cancer cells,we studied the expression pattern and function of XPLN in human colon cancer and the underlying mechanisms.In order to lay the foundation for further diagnosis and treatment of colon cancer.Method:1.The expression of XPLN in colon tumor tissues and adjacent normal tissues was detected by immunohistochemistry to explore the expression of XPLN in colon cancer cells.2.Human colon cancer HT29 cells were cultured by lentivirus transfection and divided into three groups according to the treatment methods:blank control group,negative control group and experimental group.3.The biological function of XPLN in colon cancer in vitro was investigated by establishing abnormal silencing or high expression of XPLN cell lines with the lentivirus transfection.The effects of XPLN on phosphorylation of different targets of AKT and ERK were detected as well as possible target genes.Result:We confirmed that the expression of XPLN in the adjacent tissues was higher than that in the cancer tissues.Three groups of stable HT29 cell lines were constructed for in vitro biological experiments,which showed that over-expression of XPLN in colon cancer cell is able to inhibit cell proliferation,promote cell apoptosis and reduce the ability of invasive.Further qPCR analysis of XPLN overexpressed HT29 cell lines revealed that caspase3 and VEGF may be the target genes of XPLN in colon cancer.Combined with the protein western blot,it was verified that XPLN can inhibit the phosphorylation of AKT,thus affecting the PI3K/AKT/mTOR signaling pathway,inhibiting the proliferation of colon cancer cells and promoting apoptosis ultimately.Conclusion:XPLN is low expressed in colon cancer tissues,especially in metastatic colon cancer tissues.XPLN is able to inhibit the development of colon cancer and its level is closely related to the prognosis of patients with colon cancer.The potential target genes of XPLN might include caspase3 and VEGF.Further more,it can inhibit the phosphorylation of AKT and thus inhibits the activity of related pathways.Therefor,XPLN might be a therapeutic target for diagnosis and treatment of colon cancer.
