Effect Of Extracts From Chinese Herbal Medicines On The Growth And Development Of Insulin Pathway Mutants

The disorder of carbohydrate metabolism in organisms is closely related to the occurrence of disease such as diabetes,obesity and cardiovascular.The mechanism of Drosophila regulates glucose homeostasis is highly conservative with human,and Drosophila are small,easy to operate,and composition of the system is relatively simple.Therefore,the screening of Chinese herbal medicines to alleviate diabetes by Drosophila model has gradually become a hot spot.Chinese herbal medicine has the advantages such as small toxic and side-effects,low cost and wide source.To study the effect of Chinese herbal extracts on the growth of insulin pathway mutants,the effects of 128 Chinese herbs on the growth status of flies were analyzed by the growth rate and pupal volume of mutants.The 9 kinds of Chinese herbal extracts such as Chrysanthemun indicum L.,Angelica pubescens Maxim.f.Biserrata Shan et Yuan,Cassia obtusifolia L.,Litchi chinensis Sonn.,Angelica dahurica(Fisch.Ex Hoffm.)Benth.Et Hook.f.,Portolaca oleracea L.Corydalis yanhusuo W.T.Wang,Pbyporus umbellatus(pers.)Fries andCynanchum glaucescens(Decne.)Hand.-Mazz had a mitigating effect on S6k1-1/TM6B.Moreover,the 8 kinds of Chinese herbal extracts such as Chrysanthemun indicum L.,Angelica dahurica(Fisch.Ex Hoffm.)Benth.et Hook.f.,Cynanchum glaucescens(Decne.)Hand.-Mazz,Pbyporus umbellatus(pers.)Fries,Angelica pubescens Maxim.f.Biserrata Shan et Yuan,Artemisia capillaris Thunb.,Portolaca oleracea L.and Cinnamomun cassia Presl.can improve the growth rate and the pupal volume of chico1/CyO.In study,pupal volume,body weight,wing area,fat cell size,lipid droplet and lifespan of mutants were analyzed for futher reserch.The results showed that Chrysanthemun indicum L.,Angelica pubescens Maxim.f.Biserrata Shan et Yuan,Cassia obtusifolia L.and Litchi chinensis Sonn.extracts could improve the individual size,restore the wing area,alleviate the fat storage of S6k mutants.and Angelica pubescens Maxim.f.Biserrata Shan et Yuan,Cassia obtusifolia L.and Litchi chinensis Sonn.could significantly improve the lifespan of S6k males.Among the 8 Chinese medicines screened by chico1/CyO,Chrysanthemun indicum L.,Angelica dahurica(Fisch.Ex Hoffm.)Benth.et Hook.f.,Pbyporus umbellatus(pers.)Fries,Cynanchum glaucescens(Decne.)Hand.-Mazz,Artemisia capillaris Thunb.and Cinnamomun cassia Presl.could improve the fat cell size.Futhermore,Chrysanthemun indicum L.and Cinnamomun cassia Presl.can restored the lipid droplet size of chico.The results provide basis for the alleviation and treatment of diabetes with traditional Chinese herbal medicine.
