Smilax china L.is a vine-grown plant,distributed in the area of southwest,south-central,eastern China,East Asia and Southeast Asia.It had many biological activities such as anti-inflammatory,analgesic,anti-tumor,anti-oxidation and immunosuppression.And Smilax china L.had a variety of active ingredients such as Smilax china L.alizarin,flavonoids,polysaccharides,terpenes and phenols.Polysaccharides are an importment active substance in Smilax china L.and there are few studies about it.In this study,the rhizome of Smilax china L was used as raw materials to extract the polysaccharides and the starches.The physicochemical properties,antioxidant activity,rheological properties,liver protection of SCP and gel system of SCS were initially explored.The main research contents were as follows:1.Smilax china L powder was extracted by distilled water at 95°C and SCP was obtained by removing the protein,starch,alcohol precipitation,dialysis and lyophilization.The physicochemical properties of SCP were analyzed and results showed that the total sugar content of SCP was 74.99%and the protein content was10.82%.It contained 55.28%neutral sugar and 19.71%uronic acid.The molecular weight of SCP was about 1.34×105Da,which is mainly composed of arabinose(Ara),galactose(Gal),glucose(Glc),xylose(Xyl),and galactonic acid(Gala)with a molar ratio of 2.47:7.17:34.62:10.82:1.38.The FI-IR of SCP was detected specific absorption peaks and fingerprint absorption peaks of functional groups in the range of4000-400 cm-1.When the temperature was raised to 600°C,the mass of SCP remained at 22.3%.SCP had good scavenging ability to DPPH,hydroxyl radical and superoxide anion free radical,and it had a good antioxidant activity in vitro.2.Kunming male mice(18±2g)were randomly alloted into seven groups(n=10),including normal control group,CCl4 group,biphenyl diester group(BD),low,medium and high dose group of SCP and biphenyl diester+SCP group(BS).Normal group and CCl4 group was received normal saline(0.9%NaCl,0.1 mL/10g bw)every day,and BD group was received biphenyl diester solution(200 mg/kg bw)every day.Low,medium and high doses group of SCP were daily gavaged polysaccharide solution(100,200,400mg received/kg bw SCP),and BS group was gavaged mixed solution of biphenyl diester and SCP(100 mg/kg bw biphenyl diester+100 mg/kg bw SCP).The gavage process lasted for 18 days.After last gavage for 2 h,the normal control group was intraperitoneally injected with soybean oil(0.1 mL/10 g bw).And other groups were intraperitoneally injected with 0.5%CCl4 soybean oil mixture solution(0.1 mL/10 g bw).All mice were fasted for 12 h but were free to take water in end of modeling.The blood was collected from the mice eye socket and sacrificed by cervical dislocation.The liver and spleen were dissected and determine the relevant biochemical indicators in serum and liver.The results showed that SCP can significantly reduce levels of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum by CCl4-induced(P<0.05).Compared with the CCl4 group,the medium and high dose groups of SCP could significantly increased the level of superoxide dismutase(SOD)and glutathione(GSH)and decreased the level of malondialdehyde(MDA)in the liver.The medium and high dose groups of SCP significantly reduced the level of inflammatory factors TNF-αand IL-1βin serum(P<0.05).3.The rheological properties of SCP and the effects of SCP concentration,temperature,pH and salt ion concentration on the apparen viscosity were investigated.Compared with 3%and 4%SCP,5%SCP showed good apparent viscosity,thixotropic and weak gel properties,while low temperature,low concentration of salt ion and weak acidity were beneficial to the apparent viscosity of SCP.4.The effect of Smilax China L.starch(SCS)on the gel properties and interaction of calcium ion on calcium sulfate-induced soy protein isolate gel(SPI)was investigated.The results showed that SCS had a significant effect on the gel properties and microstructure of SPI gel.With the concentration of SCS increases,the viscosity,elastic gel strength,and water holding capacity of the gel system increase.The surface of gel becomes smooth and the structure is dense.Mechanical analysis shown hydrogen bonding and disulfide interactions play a key role in gel system.
