OBJECTIVE: To establish an animal model of goat induced disc degeneration induced by controlled stress was established,and its degeneration was confirmed by imaging and pathology.MATERIALS AND METHODS: Eighteen mature goats were divided into groups: 18 goats were surgically loaded with controllable stress devices on the L4 and L5 vertebral bodies.The experimental group was a pressurized group(pressure is 40 N),and the control group was a non-pressurized group.The imaging and pathology of the goats were performed at 1,2,and 3 months after surgery.RESULTS: In the control group,the DHI measured on the X-ray was slightly decreased,but it was not statistically significant(P>0.05).The magnetic resonance T2-weighted imaging showed that the Pfirrmann grade was grade ?-?,and the HE staining showed that the nucleus pulposus and the annulus fibrosus were full.Clear,no obvious cracks.Immunohistochemical staining showed that the type ? collagen stained fiber rings were brownish yellow,which was strongly positive,and the nucleus pulposus was basically not stained.Type ? collagen fiber outer ring is basically no staining,and the nucleus pulposus is brownish yellow.In the experimental group,the DHI measured by the X-ray on the 1st,2nd and 3rd months after the operation decreased with time,which was statistically significant(P<0.05).The magnetic resonance T2-weighted imaging showed The higher the grade of Pfirrmann grading with time,???;HE staining showed that the volume of nucleus pulposus of the intervertebral disc was reduced,and the nucleus pulposus cells were reduced,which was gradually replaced by fibroid tissue.Immunohistochemical staining showed that the nucleus pulposus type ? collagen staining was light yellow?yellow?brown yellow;the nucleus pulposus type ? collagen stained from dark yellow?yellow?light yellow;suggesting that the type ? collagen of the intervertebral disc gradually increased,and the type ? collagen gradually Reduced,the disc is degenerated.Conclusion: The controllable stress device can induce the degeneration of the intervertebral disc in goats,and it will gradually aggravate with the prolongation of postoperative time.It is an ideal method for constructing the model of intervertebral disc degeneration,and the modeling operation is simple,reproducible and economical.better.
