Objective: To construct differential expression profiles of circRNA in ascending aortic middle membrane of acute type A aortic dissection(AAAD),and to preliminarily analyze the possible molecular pathogenesis of AAAD by differentially expressed circRNAs.Methods: From January 2018 to December 2018,5 cases of AAAD ascending aorta middle membrane were collected in Shenzhen sun yat-sen cardiovascular hospital;and 5 cases of donor heart transplantation ascending aorta middle membrane or aortic valve replacement ascending aortic middle membrane were collected in the same period.Screening differentially expressed circRNAs by Agilent human circRNA chip expression profiling.Relevant bioinformatics analysis using KOBAS,Kegg database and other software systems such as miRanda,Cytoscape,preliminary prediction of AAAD possible signaling pathways and regulatory mechanisms.Results: 1.Through inter-group comparison,there were 588 differentially expressed circRANs in AAAD group and control group,363 differentially expressed circRANs were up-regulated,and 225 differentially expressed circRANs were down-regulated.2.GO analysis showed that the differentially expressed circRNAs in the AAAD group involved 31 cell components,including extracellular space,laminin-1 complex and so on;291 biological processes,including cell activation,regulation of cell adhesion and so on;30 molecular functions,including receptor signaling protein serine/threonine kinase activity,protein tyrosine phosphatase activity,etc.3.Kegg pathway analysis showed that differentially expressed circRNAs target genes were involved in a total of 22 signaling pathways,among which MAPK signaling pathway and mTOR signaling pathway were significantly enriched.4.circRNA/miRNA binding site analysis revealed that 588 circRNAs differentially predicted to bind more than 10,000 miRNAs,including 30 aortic dissection-related miRNAs,such as has-miR-149-5p,has-miR-146b-3p,etc.In the circRNA/miRNA interaction network,these 30 aortic dissection-associated miRNAs bind to 179 differentially expressed circRNAs,of which has-miR-149-5p,predicted to bind 43 differentially expressed circRNAs,of which has-circ-0017177,predicted to combine 18 aortic dissection pathologically relevant miRNAs.Conclusion: 1.Compared with the control group,there were a large number of differentially expressed circRANs in the middle membrane of the ascending aortic wall in group AAAD.These differentially expressed circRNAs may be closely related to the occurrence and development of acute type A aortic dissection.2.GO analysis showed that differential expression of circRNAs may involve physiological and pathological processes such as cell activation,proliferation and apoptosis,exogenous matrix synthesis and degradation,and cell adhesion.3.MAPK signaling pathway and mTOR signaling pathway may be closely related to the occurrence and development of AAAD.4.circRNA/miRNA binding site analysis suggests that the interaction mechanism between circRNA and miRNA is complex,and it is not a one-to-one correspondence.The specific regulatory mechanism remains to be further explored.
