Purpose:To investigate the expression of PD-1 and its ligand PD-L1 in cervical cancer tissues, and to analyze the relationship between PD-1/PD-L1 expression and the clinicopathological features of cervical cancer. Methods : 1.8 chronic cervicitis tissues and 28 cervical squamous intraepithelial lesion tissues and 56 tissues of cervical cancer were collected.2.The expression of PD-1 and PD-L1 protein in normal cervical, cervical squamous intraepithelial lesion tissues(SIL)and cervical cancer tissues were detected by immunohistochemistry.3.To analyze the relationship between the expression of PD-1/PD-L1 in cervical cancer tissues and clinicopathological features.Results:1.The PD-1 protein was expressed in small amounts in the cervical tissues of chronic cervicitis, and its expression level was lower than that in LSIL tissues, which was significantly lower than that in the HSIL tissues and cancer tissues. The expression of PD-1 was significantly up-regulated in cervical cancer tissues, and the high expression rate was 76.79%.The expression of PD-1 in cervical cancer tissues was significantly higher than that in PD-1 in low-grade SIL tissues and highgrade SIL tissues.2.The PD-L1 protein was expressed in a small amount in the cervical tissues of chronic cervicitis, and its expression level was lower than that in the LSIL tissue, which was significantly lower than that in the HSIL tissues and cervical cancer tissues. The high expression rate of PD-L1 in cervical cancer tissues was 62.50%.With the progress of cervical tissue lesions, the expression level of PD-L1 increased gradually in LSIL tissues,HSIL and cervical cancer tissues.3.The two-dimensional distribution map found that PD-1 and PD-L1 have certain regional distribution characteristics in different groups of samples. We can see that the chronic cervicitis can be distinguished from the cervical cancer and HSIL tissue obviously.Meanwhile the chronic cervicitis can be distinguished from the LSIL tissue basically, but cervical cancer and high-grade SIL tissues cannot be better differentiated.4.There was no significant correlation between PD-1/PD-L1 expression and age,FIGO stage, tumor differentiation, and HPV infection.Conclusions:1.The expression levels of PD-1 and PD-L1 in chronic cervicitis tissues are extremely low,and they are expressed at low levels in LSIL tissues, but elevated expression in high-grade SIL tissues and cervical cancer tissues. It suggests that PD-1 and PD-L1 are related to the occurrence and development of cervical lesions;it may be one of the early tumor markers of cervical cancer.2.There was no significant correlation between PD-1/PD-L1 expression and age,FIGO stage, tumor differentiation, and HPV infection.
