Objective: This study explored the effect of PKM2 expression on proliferation and invasion of osteosarcoma at the cellular level,providing a theoretical basis for PKM2 to become a potential target for early diagnosis,treatment and prognosis of osteosarcoma.Methods: 1.Osteosarcoma cells MG63 transfected with si RNA-PKM2 were used as the experimental group,while the MG63 cells in normal culture and transfected with unrelated sequences were used as the control group.2.Western blot was used to detect the expression changes of PKM2.3.CCK-8 assay and plate cloning assay were used to detect the effect of reducing PKM2 on the proliferation capacity of osteosarcoma cell MG63.4.Transwell assay was performed to detect the effect of reducing PKM2 on the invasion ability of osteosarcoma cell line MG63.Results:1.Expression of PKM2 were significantly decreased in MG63 cells transfected with si RNA-PKM2,and the difference was statistically significant(P < 0.05)compared with the control group of normal cultured cells and transfected unrelated sequences.2.In CCK-8 proliferation test,the absorbance value of MG63 cells transfected with si RNA-PKM2 at 48 h and 72 h was significantly lower than that of the control group(P <0.05).3.In the plate cloning experiment,the clone count of MG63 cells transfected with si RNA-PKM2 was 79.34±5.01,significantly less than the control groups(P < 0.05).4.In the Transwell experiment,the number of transmembrane cells of MG63 cells transfected with si RNA-PKM2 was 82.00±11.72,which was statistically significant compared with the control group(P<0.05).Conclusions: Reducing of PKM2 expression significantly decreased the proliferation and invasion ability of osteosarcoma cells,indicating that PKM2 can promote the proliferation and invasion of osteosarcoma cells.
