OBJECTIVE: To study the effect of metastasis associated in colon cancer1(MACC1)on malignant biological behavior of human nasopharyngeal carcinoma cells,and to observe the effect of overexpression of MACC1 on proliferation,invasion and migration of human nasopharyngeal carcinoma cells.Methods: 1.To detect the protein expression levels of MACC1 in 5-8F?HNE-1?CNE-1 and CNE-2 cell lines of nasopharyngeal carcinoma by Western blot.2.To establish a group of CNE-2 cell lines with overexpression of MACC1 by lentivirus transfection,and to select cells with stable transfection of MACC1.3.The experiment was divided into three groups: experimental group(CNE-2 MACC1 overexpression stable strain group);negative control group(CNE-2 transfection empty vector group);blank control group(CNE-2 empty cell group).4.To detect the expression level of MACC1 in three groups of cells by Q-PCR and the Western blot.5.To observe the proliferation ability among cells in each group by MTT assay.6.To observe the migration ability among cells in each group by Transwell migration experiment.7.To observe the invasive ability among cells in each group by Transwell invasion experiment.Res?lts:1.1.Western blot results showed that the highest expression level of MACC1 was found in HNE-1 cell line,and moderate in CNE-1cell line,and was not detected or very low in CNE-2 and 5-8F cell lines.2.After lentivirus transfection,green fluorescence expression was observed in the experimental group cells under inverted fluorescence microscope.3.Q-PCR results showed that the expression of MACC1 in the experimental group was much higher than that in the negative control group and the blank control group.4.The Western blot results showed that the cells in the experimental group had obvious bands and MACC1 was successfully expressed,while the cells in the control group did not have bands,suggesting that MACC1 had no expression or extremely low expression.5.MTT results showed that the proliferative ability of the cells in the experimental group was significantly higher than that of the control group after 24 h,48h,72 h of lentivirus transfection,there was significant difference between the experimental group and the control group(P<0.05).6.The results of Transwell migration experiment showed that after24 hours of culture,the migration number of cells in the experimental group was(85±4),that in the negative control group was(59±3),and that in the blank control group was(58±4),and the difference between the experimental group and the control group was statistically significant(P<0.05).7.Transwell invasion experiment showed that after 24 hours of culture,the number of cells in the experimental group was(81±4),in the negative control group was(42±3)and in the blank control group was(40±4).There was significant difference between the experimental group and the control group(P<0.05).Conclusion: The abnormal high expression of MACC1 will lead to the enhancement of malignant biological behaviors such as proliferation,migration,and invasion of nasopharyngeal carcinoma CNE-2 cells,suggesting that MACC1 has participated in the process of tumor development such as procreation,invasion,and metastasis of nasopharyngeal carcinoma.
